Here we report the 1,3-dipolar cycloaddition of polycyclic aromatic azomethine ylides with nitriles to produce highly fused imidazole derivatives, that is, tribenzo[b,g,ij]imidazo-[2,1,5-de]quinolizine. The advantages of this transformation are the broad substrate scope and the good functional group compatibility. The subsequent palladium-catalyzed intramolecular cyclization provides an efficient approach to further pi-extended imidazoles, that is, 14b(1),15-diazadibenzo[fg,ij]cyclopenta[rst]pentaphene.

One hot debate between catalytic activities of sulfonated reduced graphene oxide (RGO-SO3H), as the 2D heterogeneous carbon-based catalyst, and sulfonated multi-walled carbon nanotubes (MWCNTs-SO3H), as the 1D heterogeneous carbon-based catalyst, was investigated in the synthesis of 1,3,5-triarylbenzenes under different conditions. This comparison study revealed that the 2D catalytic system was more efficient relative to the 1D catalyst in terms of yields of the target products, turnover frequency of the catalyst (TOF), and the reusability. The reasons of this observation such as specific surface area, confinement spaces in 1D and 2D carbon-based catalysts, solvent accessible surface area, surface active sites, and the availability of supported functional groups on carbon nanostructures will discuss. Moreover, the application of synthesized compounds as a substrate was checked in carbon-carbon bond formation. Also, one of the synthesized compounds was produced using three approaches under metal and non-metal conditions. Notably, the recyclability of the two catalytic systems was checked.

alpha-Glucosidase inhibition has been considered as an effective strategy for controlling the post-prandial hyperglycemia in diabetic patients. A series of novel isatin-pyrazole hybrids has been rationally designed, synthesized and characterized by different spectroscopic techniques. All of the synthesized compounds were evaluated for their alpha-glucosidase inhibitory potential using in vitro enzymatic assay. The tested compounds have exhibited potent alpha-glucosidase inhibition with IC50 values ranging from 3.26 +/- 0.25 to 32.33 +/- 1.08 mu M. Compound V7, having 3-nitro phenyl ring appended at pyrazole nucleus, has emerged as the most potent inhibitor among the series with an IC50 value of 3.26 +/- 0.25 mu M, which was approximately 146 fold more potent than the clinically used drug acarbose (IC50 = 478.07 +/- 1.53 mu M). Molecular docking studies were carried out to investigate the binding interactions of most active compounds with the active site of the enzyme. Further, in silico predictive pharmacokinetic parameters (ADME) and drug-like properties of the compounds were within the acceptable ranges. In vivo biological evaluations further supplemented the in vitro results.

Several commercially available lipases were examined in a study on O-Si bond formation and cleavage applying silicon-based protecting groups and alcohols or the corresponding silyl ethers. With regard to deprotection, from silyl ether to the corresponding alcohol, only the solvent and the lipase were necessary. Zr-based metal-organic frameworks have been considered promising heterogeneous catalysts in various reactions. It has been proven that the defect sites in these frameworks, which result from deficiency of linkers, could act as the reaction active sites. In this work, microwave irradiation was used for rapid synthesis and increasing defects on Zr-6 clusters of metal-organic framework UiO-66. Powder X-ray diffraction, thermal gravimetric analysis, and N-2 sorption isotherms were performed to characterize and calculate the defect number in the framework. UiO-66 prepared by microwave method for a few minutes could reach 1.8 missing linkers per cluster. The material with acetic acid as a co-catalyst showed highly heterogeneous catalytic activity in the solvent-free heterocyclization of 2,2-disubstituted 2,3-dihydroquinazolin-4(1H)-ones from anthranilamide and ketones/aldehydes. The synthetic scheme consisted of zirconium cluster catalysed carbonyl group of anthranilamide to achieve the C-N propitious combination and succeeding ring closure process.

The use of ubiquities elements such as iron instead of expensive precious metals as catalysts is one goal toward realizing environmentally benign synthetic chemistry. Here, we report that porous FeO(OH) dispersed on Mg-Al hydrotalcite acts as a bifunctional heterogeneous catalyst in the one-pot synthesis of 2-substituted quinoline derivatives through dehydrogenative oxidation-cyclization reactions. The catalyst was prepared by a simple grafting method using FeCl3 and Mg-Al hydrotalcite. The prepared porous FeO(OH) possesses a higher surface area than those previously reported for alpha-FeO(OH) particles. The one-pot quinoline synthesis proceeded effectively under non-noble-metal catalysis in air without requiring additional homogeneous bases or solvents.

Y By removing 5-methyl and 6-acetyl groups in our previously reported compound 3, we designed a series of novel 2,7-diaryl-[1,2,4]triazolo[1,5-a]pyrimidine derivatives as potential tubulin polymerization inhibitors. Among them, compound 5e displayed low nanomolar antiproliferative efficacy on HeLa cells which was 166-fold higher than the lead analogue 3. Interestingly, 5e displayed significant selectivity in inhibiting cancer cells over HEK-293 (normal human embryonic kidney cells). In addition, 5e dose-dependently arrested HeLa in G2/M phase through the alterations of the expression levels of p-cdc2 and cyclin B1, and caused HeLa cells apoptosis by regulation of expressions of cleaved PARP. Further evidence demonstrated that 5e effectively inhibited tubulin polymerization and was 3-fold more powerful than positive control CA-4. Moreover, molecular docking analysis indicated that 5e overlapped well with CA-4 in the colchicine-binding site. These studies demonstrated that 2,7-diaryl-[1,2,4]triazolo [1,5-a]pyrimidine skeleton might be used as the leading unit to develop novel tubulin polymerization inhibitors as potential anticancer agents.
