The cationic organotin cluster [t-Bu2Sn(OH)(H2O)](2)(2+)2OTf(-) is easy to prepare and stable in air. The catalytic activity of [t-Bu2Sn(OH)(H2O)](2)(2+)2OTf(-) as a neutral organotin Lewis acid catalyst is probed through the one-pot three-component syntheses of 5-substituted 1H-tetrazoles from aldehydes, hydroxylamine hydrochloride and sodium azide, and of 2,4,6-triarylpyridines from aromatic aldehydes, substituted acetophenones and ammonium acetate. The reactions proceed well in the presence of 1 mol% of [t-Bu2Sn(OH)(H2O)](2)(2+)2OTf(-) in water and provide the corresponding 5-substituted 1H-tetrazoles and 2,4,6-triarylpyridines in good to excellent yields. The method reported has several advantages such as the catalyst being neutral, low catalyst loading and use of water as a green solvent.

Novel substituted bis-(3-cyano-2-pyridone) derivatives were prepared via a powerful method using enaminonitriles push-pull dienes as key building blocks. The synthesis was performed in three steps from easily accessible starting materials in good yields. All target products were structurally elucidated by spectroscopic data. The reaction mechanism for formation of bis (3-cyano-2-pyridone) derivatives was proposed. The structures of enaminonitriles 3a and bis-pyridones 4c were confirmed by single-crystal X-ray diffraction. The florescent study of some bis (3-cyano-2-pyridones) derivatives was reported. Many natural or synthetic chalcones have potential anti-tumor activity. Here, we synthesized two series of chalcone analogues containing a thieno [2,3-cflpyrimidin-2-y] group and evaluated for their cytotoxic activity towards cultured human lung cancer A549 and colorectal HCT-116 cells. Among them, compound 8d was the most cytotoxic against HCT-116 cells, with an IC50 value of 2.65 mu M. Analyses of the phenotypic changes induced by this compound found a dose-dependent accumulation of HCT-116 cells in sub-G1 phase, indicating that compound 8d might induce apoptosis. Furthermore, we found that 8d triggered mitochondrial membrane potential depolarization, promoted reactive oxygen species formation in HCT-116 cells, and increased the percentage of early and late apoptotic cells. Finally, immunoblotting indicated that 8d increased PARP-1 and caspases 3, 7 and 9 cleavage. These data suggest that compound 8d induces apoptosis via the mitochondrial death pathway.

Coordination of 1,4-disubstituted 1,2,3-triazoles L-1 and L-2 with [(p-cymene)RuCl2](2) followed by dehydrochlorination in the presence of a base resulted in the formation of complexes 1 and 2, respectively. Both were tested for the transfer hydrogenation of aldehydes and ketones in air using ecologically benign and cheap ethanol as the hydrogen source in the presence of a catalytic amount of a base. Air-stable complex 1 was proved to be an active catalyst for the transfer hydrogenation of a wide variety of aromatic and aliphatic aldehydes and ketones bearing various functionalities. Catalyst 1 was also effective for the transfer hydrogenation of carbonyls using the simplest primary alcohol, methanol, under aerobic conditions. Under the present catalytic protocol, labile or reducible functionalities such as nitro, cyano, and ester groups were tolerated. Good selectivity was also observed for acyclic alpha,beta-unsaturated carbonyls. However, this catalytic protocol was not selective for 2-cyclohexen-1-one as both alkene and keto moieties were reduced. The transfer hydrogenations are believed to proceed via a ruthenium-hydride intermediate. Finally, transfer hydrogenation of acetophenone using isopropanol as a commonly used hydrogen source was also performed and the sustainable and green credentials of these catalytic protocols utilizing methanol, ethanol, and isopropanol were compared with the help of the CHEM21 green metrics toolkit.

A strategy for heterogeneous copper(I) mesoporous silica [Cu(I)-SBA-15] mediated catalytic reduction has been developed for the reduction of various nitroarenes in excellent yields. In addition to alcothermal strategy, other strategies have been attempted for the preparation of Cu(I) substituted mesoporous silica, however, the desired oxidation state is obtained only through alcothermal strategy. The crystalline phases and oxidation state of synthesized materials were characterized by XRD and XPS measurements and tested for catalytic reduction of nitroarenes. Among the tested materials, the best catalytic reduction of nitroarenes with an excellent substrate scope and good catalytic reusability was established by Cu(I)-SBA-15 material. The reduction kinetics and the controlled experimental studies reveal that the reaction proceeds through pseudo-first-order kinetics and nitrosobenzene intermediate pathway.

Chiral ionic liquids (CILs) have shown a wide range of applications in variety of domains in chemistry. Because of this, synthesis and applications of CILs have always been areas of interest for research in the last 20 years. Present work describes, the synthesis of six carbohydrate based chiral ionic liquids (CCILs) by following simple procedures and their applications. Structures of the cats were confirmed through various analytical techniques like NMR spectroscopy (H-1, C-13, B-11, P-31,F-19), El-MS, and polarimetry. Designed CCILs were tested as chiral recognising agents using sodium salt of Mosher’s acid as model substrate through (FNMR)-F-19-N- spectroscopy. Hollow magnetic nanoparticles (MNPs) with tetrahedral morphology were synthesized and then covered by a shell prepared by coating with melamine-formaldehyde followed by the introduction of glucose-derived carbon. Subsequently, Pd nanoparticles were immobilized and the core-shell nanocomposite was carbonized. The obtained magnetic catalyst was successfully applied for the hydrogenation of nitroarenes in aqueous media. To investigate the effects of the morphology of MNPs, the nature of carbon shell, and the order of incorporation of Pd nanoparticles, several control catalysts, including the MNPs with different morphologies (disc-like and cylinder); MNPs coated with different shells (sole glucose-derived carbon or melamine-formaldehyde carbon shell); and a nanocomposite, in which Pd was immobilized after carbonization, were prepared and examined as catalyst for the model reaction. To justify the observed different catalytic activities of the catalysts, their Pd loadings, leaching, and specific surface areas were compared. The results confirmed that tetrahedral MNPs coated with porous N-rich carbon shell exhibited the best catalytic activity. The high catalytic activity of this catalyst was attributed to its high surface area and the interaction of N-rich shell with Pd nanoparticles that led to the higher Pd loading and suppressed Pd leaching.

Chiral, cationic NHC-iridium complexes are introduced as catalysts for the intramolecular hydroamination reaction of unactivated aminoalkenes. The catalysts show high activity in the construction of a range of 5- and 6-membered N-heterocycles, which are accessed in excellent optical purity, with various functional groups being tolerated with this system. A major deactivation pathway is presented and eliminated by using alternative reaction conditions. A detailed experimental and computational study on the reaction mechanism is performed providing valuable insights into the mode of action of the catalytic system and pointing to future modifications to be made for this catalytic platform.

Leishmaniasis is considered as one of the major neglected tropical diseases due to its magnitude and wide geographic distribution. Leishmania braziliensis, responsible for cutaneous leishmaniasis, is the most prevalent species in Brazil. Superoxide dismutase (SOD) belongs to the antioxidant pathway of the parasites and human host. Despite the differences between SOD of Leishmania braziliensis and human make this enzyme a promising target for drug development efforts. No medicinal chemistry effort has been made to identify LbSOD inhibitors. Herein, we show that thermal shift assays (TSA) and fluorescent protein-labeled assays (FPLA) can be employed as primary and secondary screens to achieve this goal. Moreover, we show that thiazole derivatives bind to LbSOD with micromolar affinity.
