An efficient and facile approach has been described for the synthesis of a new series of 1-amino-2-oxo-nicotinonitriles and their sulfonamides under microwave and ultrasonic irradiation. The desired compounds 4 a-l were obtained via condensation of p-chlorobenzaldehyde, aryl or heteroaryl methyl ketone and cyanoacetohydrazide in the presence of catalytic piperidine under three synthetic methodologies (namely, conventional method, ultrasonic and microwave irradiation), the best results (short reaction times, pure products, high yield) were obtained by ultrasonic irradiation. Sulfonamides 5 a-f resulted via reaction of amines 4 a-e and 4 j with benzene sulfonyl chloride in the presence of pyridine. The synthesized compounds give no significant results when investigated against Gm (+ve) and Gm (-ve) bacteria, and fungi strains.

The secondary amine participating asymmetric reductive amination remains an unsolved problem in organic synthesis. Here we show for the first time that secondary amines are capable of effectively serving as N-sources in direct asymmetric reductive amination to afford corresponding tertiary chiral amines with the help of a selected additive set under mild conditions (0-25 degrees C). The applied chiral phosphoramidite ligands are readily prepared from BINOL and easily modified. Compared with common tertiary chiral amine synthetic methods, this procedure is much more concise and scalable, as exemplified by the facile synthesis of rivastigmine and N-methyl-1-phenylethanamine.

A sustainable synthesis of sulfonamides using a copper-catalysed process starting from triarylbismuthines, Na2S2O5 and nitro compounds in a Deep Eutectic Solvent (DES) as a reaction medium is described. Thus, triarylbismuthines are used as reagents for the incorporation of SO2 into organic motifs. The bismuth salts formed as by-products can be easily removed from the crude reaction mixture by precipitation with water, while the use of volatile organic compounds (VOCs) as solvents can be avoided in the entire process. The eutectic mixture employed as the solvent is fully characterised, with the preliminary results proving its low toxicity. The designed DES also allows for a novel multicomponent reaction which saves time and reduces purification steps, energy and cost.

The regioselective N-addition/substitution reaction between alpha-alkylidene pyrazolinones and propargyl sulfonium salts has been developed to construct functionalized allylthio-containing pyrazolones with moderate to excellent yields. alpha-Alkylidene pyrazolinones act as N-nucleophilic agents which are distinguished from reported C-nucleophilic reactions. alpha-Glucosidase inhibition has been considered as an effective strategy for controlling the post-prandial hyperglycemia in diabetic patients. A series of novel isatin-pyrazole hybrids has been rationally designed, synthesized and characterized by different spectroscopic techniques. All of the synthesized compounds were evaluated for their alpha-glucosidase inhibitory potential using in vitro enzymatic assay. The tested compounds have exhibited potent alpha-glucosidase inhibition with IC50 values ranging from 3.26 +/- 0.25 to 32.33 +/- 1.08 mu M. Compound V7, having 3-nitro phenyl ring appended at pyrazole nucleus, has emerged as the most potent inhibitor among the series with an IC50 value of 3.26 +/- 0.25 mu M, which was approximately 146 fold more potent than the clinically used drug acarbose (IC50 = 478.07 +/- 1.53 mu M). Molecular docking studies were carried out to investigate the binding interactions of most active compounds with the active site of the enzyme. Further, in silico predictive pharmacokinetic parameters (ADME) and drug-like properties of the compounds were within the acceptable ranges. In vivo biological evaluations further supplemented the in vitro results.

Two cis-1,2-diol-type chiral ligands (T-1 and T-2) and their tri-coordinated chiral dioxaborinane (T(1-2)B(1-2)) and four-coordinated chiral dioxaborinane adducts with 4-tert-butyl pyridine sustained by N -> B dative bonds (T(1-2)B(1-2)-N) were synthesized and characterized by various spectroscopic techniques such as NMR (H-1, C-13, and B-11), FT-IR and UV-Vis spectroscopy, LC-MS/MS, and elemental analysis. Then, all chiral dioxaborinane complexes were investigated as catalysts for the asymmetric transfer hydrogenation of various ketones under suitable conditions. We demonstrate a method to boost the photocatalysis of titania by constructing a core-shell structure. The crucial technique is to capsulate the titania nanoparticles by a sheath of nitrogen-doped carbon (TiO2@N-AC) with a thickness at 3.5-7.5 nm, which creates an exceptional effect to accelerate the catalytic hydrogenation of substituted nitroarenes with isopropanol as the hydrogen source under ultraviolet or visible-light irradiation. In the catalytic hydrogenation of various nitroarenes with substituted groups of -Cl, -F, -C-C, -C-O, and -C=N, TiO2@N-AC exhibits high conversions and selectivities to the corresponding functionalized anilines, even outperforming the catalysts containing noble metals. Mechanism study demonstrates that the TiO2@N-AC catalyst benefits the selective adsorption of the nitro group rather than other substituted groups, which efficiently avoids the undesirable side reactions. Importantly, the catalyst is stable and exhibits constant performances in the continuous recycling tests.

The ketones was successfully prepared from secondary alcohols using 9-azabicyclo[3.3.1]nonane-N-oxyl (ABNO) as the catalyst and 2,6-lutidine as the base in acetonitrile solution. The electrochemical activity of ABNO for oxidation of 1-phenylethanol was investigated by cyclic voltammetry, in situ Fourier transform infrared spectroscopy (FTIR) and constant current electrolysis experiments. The resulting cyclic voltammetry indicated that ABNO exhibited much higher electrochemical activity when compared with 2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) under the similar conditions. A reasonable reaction mechanism of the electrocatalytic oxidation of 1-phenylethanol to acetophenone was proposed. In addition, a series of secondary alcohols could be converted to the corresponding ketones at room temperature in 80-95% isolated yields.
