The title compounds, 3,3′,3.,3”’-[methylenebis(oxybenzene-5,1,3-triyl)]tetrakis(6,8-diaryl[1,2,4]triazolo[3,4-b][1,3,4]thiadiazepines), were synthesized starting from dimethyl 4-hydroxyisophthalate which was reacted with dibromomethane to obtain tetramethyl 5,5′-[methylenebis(oxy)]di(benzene-1,3-dicarboxylate). The latter was treated with excess hydrazine hydrate, and the resulting tetrahydrazide was converted to tetrakis1,2,4-triazolyl derivative via cyclization with ethanolic potassium hydroxide, carbon disulfide, and hydrazine hydrate. In the final stage, cyclocondensation with substituted chalcones afforded the target products. The structure of the newly synthesized compounds was confirmed by elemental analyses and FT-IR and NMR spectra, and their antimicrobial activities against some bacterial and fungal strains were estimated by the disc diffusion method.

A new metal-, oxidant-, and solvent-free ecofriendly domino method has been established for modular synthesis of a diverse range of medicinally promising hydroxyarylated unsymmetrical pyridines in good to high chemical yields with an excellent regioselectivity. This domino process involves a range of N-sulfonyl ketimines as C,N-binucleophiles, enolizable ketones, and aromatic/heteroaromatic aldehydes using ammonium acetate as an ideal promoter under neat conditions, which creates two new C-C bonds and one C-N bond. Notably, the neutral reaction conditions are mild enough to tolerate a range of functionalities and cover a variety of substrates, thus bestowing a powerful avenue to access tri- and tetrasubstituted pyridines including carbo- and heterocyclic fused ones. Interestingly, a practical, scalable, and high-yielding synthesis of pyridylphenol derivatives was successfully accomplished by our unique method.

This work deals with the synthesis, spectroscopic studies and catalytic evaluation of the novel chiral salen (L1H2) and (L2H2) ligands and their chiral boronate [L-1(B1-4)] and [L-2(B1-4)] complexes. Initially, the reaction of 5-azidomethyl salicylaldehyde and (R)-(-)-2-amino-1-butanol in absolute ethanol afforded a new chiral salen ligand (L1H2). Then, a novel chiral salen ligand (L2H2) have been prepared from chiral salen ligand (L1H2) for the synthesis of boronate [L-2(B1-4)] complexes through click reaction approach under ambient conditions. The reaction of chiral salen (L1H2) and (L2H2) ligands with various boronic acids afforded a new tetra-coordinated mononuclear chiral boronate [L-1(B1-4)] and [L-2(B1-4)] complexes. All the compounds are remarkably stable crystalline solids and were obtained in good yields. For the full characterization of newly synthesized chiral salen ligands and their boronate complexes, the FT-IR, UV-Vis, NMR (H-1, C-13, and B-11), LC-MS, and elemental analysis techniques have been used. The well-shaped chiral boronate compounds were investigated as catalyst for the asymmetric transfer hydrogenation (ATH) of aromatic ketones under appropriate settings. Particularly, it was proved that the ferrocene-based boronate compounds can afford an efficient catalytic conversion compared to the other boronate complexes in the asymmetric transfer hydrogenation catalytic studies. Protein disulfide isomerase (PDI) is responsible for nascent protein folding in the endoplasmic reticulum (ER) and is critical for glioblastoma survival. To improve the potency of lead PDI inhibitor BAP2 ((E)-3-(3-(4-hydroxyphenyl)-3-oxoprop-1-en-1-yl)benzonitrile), we designed and synthesized 67 analogues. We determined that PDI inhibition relied on the A ring hydroxyl group of the chalcone scaffold and cLogP increase in the sulfonamide chain improved potency. Docking studies revealed that BAP2 and analogues bind to His256 in the b’ domain of PDI, and mutation of His256 to Ala abolishes BAP2 analogue activity. BAP2 and optimized analogue 59 have modest thiol reactivity; however, we propose that PDI inhibition by BAP2 analogues depends on the b’ domain. Importantly, analogues inhibit glioblastoma cell growth, induce ER stress, increase expression of G2M checkpoint proteins, and reduce expression of DNA repair proteins. Cumulatively, our results support inhibition of PDI as a novel strategy to treat glioblastoma.

Graphitic carbon nitride (g-C3N4) and graphene (GO) have been greatly utilized as supports in the field of heterogeneous catalysis. In this work, layered C3N4 polymer/graphene hybrid (CNNS/rGO(20)) with heterostructure was fabricated by a hydrothermal method followed by loading Pd nanoparticles on the hybrid. The palladium was well dispersed uniformly (1.31 nm) owing to the layered and porous heterostructure of CNNS/rGO(20). The obtained catalyst was used for the transfer hydrogenation of a series of nitro-compounds to give the corresponding aromatic amines with outstanding activity by employing formic acid as hydrogen donor under mild conditions. The catalytic activity of the heterogeneous catalyst showed no significant loss after five continuous use.

In order to introduce the concept of covalent organic frameworks (COFs), an important class of predictable crystalline porous polymers, an integrated laboratory experiment for advanced organic chemistry students is reported. The importance of these kind of polymers and their multiple applications are presented as a part of key concepts in polymer and organic chemistry, bridging these disciplines to materials science. Students carry out the synthesis and characterization of an imine-linked COF and its building blocks. The polymer is formed at room temperature by using solvent-assisted synthesis and mechanochemistry. Students are trained in solid state characterization techniques, including FTIR spectroscopy, thermogravimetric analysis (TGA), C-13 NMR cross-polarization-magic angle spinning (CP-MAS) NMR spectroscopy, and powder X-ray diffraction (PXRD), which are essential in the important field of reticular chemistry. Therefore, this lab experience constitutes a practical introduction to COFs.

A novel nonanuclear copper(II) complex obtained by a facile one-pot self-assembly was found to catalyze the hydroboration of ketones and aldehydes with the absence of an activator under mild, solvent-free conditions. The catalyst is air-and moisture-stable, displaying high efficiency (1980 h(-1) turnover frequency, TOF) and chemo-selectivity on aldehydes over ketones and ketones over imines. This represents a rare example of divalent copper catalyst for the hydroboration of carbonyls.

Glutathione (GSH), the most abundant nonprotein thiol found in living organisms, are involved in the etiology and progression of many human diseases including cancer. So, monitoring changes of cellular GSH levels has an important guiding significance. To date, however, majority of probes can only qualitatively detect GSH in living cells. Herein, with coumarin as the read-out fluorophore and Michael addition as the sensing mechanism, six fluorescent probes were designed and synthesized. Among them, RP-2 exhibited a reversible and extremely fast response toward GSH (half time: similar to 3 s), which endowed RP2 the capacity of real-time imaging. Among the reversible probes based on Michael addition, RP-2 had both the largest forward and reverse rate constants thus far. The reaction between RP-2 and GSH was studied in detail by density functional theory and fluorescence spectroscopy. Real-time imaging of GSH levels in living cells was achieved with a temporal resolution of seconds. To simplify the processing of images, a program was developed and validated. RP-2 was expected to serve as a new fluorescent imaging tool to understand the function of intracellular GSH in the future.

A useful and efficient approach for the synthesis of nitroarenes from several aromatic amines (including heterocycles) using peroxide and base has been developed. This oxidative reaction is very easy to handle and afforded the products in good yields.
