An efficient one pot strategy has been developed for the synthesis of 2-acylbenzothiazoles and quinoxalines from aryl methyl ketones. A variety of heterocycles of these two categories could be prepared in a two-step sequence in excellent yields. The reaction was performed starting from aromatic ketones via an aryl gloxal intermediate which, on further condensation with 2-aminobenzenethiol or o-phenylenediamine, produces the corresponding heterocyclic skeleton. Aromatic ketones are initially treated with TsNBr2 in DMSO at 65 degrees C for 3 h and the crude reaction mixture was treated with 2-aminobenzenethiol (at 80 degrees C) or o-phenylenediamine (at RT) to generate the final compound.

Alzheimer’s disease is a degenerative brain condition that is the leading cause of dementia affecting millions of people around the world. Therapeutic development has focused on the problem of the loss of basal forebrain cholinergic function, as it is the only evidence responsible for brain neurodegeneration in patients with Alzheimer’s disease. Several attempts to improve cholinergic neurotransmission have been investigated by minimizing synaptic degradation of acetylcholine using acetylcholinesterase inhibitors. In the current study, we explore the designing of a new series of nicotinonitrile-coumarin hybrids as potential acetylcholinesterase inhibitors. The new hybrids were prepared utilizing pyridine-2(1H)-thiones as starting precursors. The in vitro acetylcholinesterase (AChE) inhibitory activities were examined for the new nicotinonitrile-coumarin hybrid molecules, when compared with donepezil as a standard drug with IC(50)of 14 nM. Coumarin derivative, linked to 6-(4-nitrophenyl)-4-phenylnicotinonitrile, showed more effective inhibitory activity than the reference donepezil with IC(50)of 13 nM. The free radical-scavenging capabilities against DPPH of the new hybrid derivatives were screened. Additionally, their in vitro cytotoxic activities have been tested against various eukaryotic cells. Furthermore, docking study showed excellent interaction between nicotinonitrile-coumarin hybrids and AChE. [GRAPHICS] .

We prepared high-performance aromatic copolyamides, containing bithiazole and thiazolo-thiazole groups in their main chain, from aromatic diamines and isophthaloyl chloride, to further improve the prominent thermal behavior and exceptional mechanical properties of commercial aramid fibers. The introduction of these groups leads to aramids with improved strength and moduli compared to commercialmeta-oriented aromatic polyamides, together with an increase of their thermal performance. Mono- or di-substituted imidazole derivatives were synthesized using a one-pot, two-step strategy. All imidazole derivatives were tested for AChE and BChE inhibition and showed nanomolar activity similar to that of the test compound donepezil and higher than that of tacrine. Structure activity relationship studies, docking studies to on X-ray crystal structure of AChE with PDB code 1B41, and adsorption, distribution, metabolism, and excretion (ADME) predictions were performed. The synthesized core skeleton was bound to important regions of the active site of AChE such as the peripheral anionic site (PAS), oxyanion hole (OH), and anionic subsite (AS). Selectivity of the reported test compounds was calculated and enzyme kinetic studies revealed that they behave as competitive inhibitors, while two of the test compounds showed noncompetitive inhibitory behavior. ADME predictions revealed that the synthesized molecules might pass through the blood brain barrier and intestinal epithelial barrier and circulate freely in the blood stream without binding to human serum albumin. While the toxicity of one compound on the WS1 (skin fibroblast) cell line was 1790 mu M, its toxicity on the SH-SY5Y (neuroblastoma) cell line was 950 mu M.

This work presents a highly active and reusable heterogeneous film catalytic assembly for hydrogenation reduction of aromatic nitro compounds. The multilayer structures of PEI-(K2PdCl4-P1)(n)-film (PEI = polyethylenmine, P1 = 3-amino-3-(4-pyridinyl)-propionitrile) bound to quartz slides were fabricated by layer-by-layer (LbL) self-assembly method. Various characterization techniques including X-ray photoelectron spectroscopy (XPS), inductively coupled plasma OES spectrometer (ICP), UV-vis spectroscopy and atomic force microscopy (AFM) were employed to reveal the growth process of the resulting LbL multilayers in detail. Subsequent in situ reduction by H-2 produced Pd nanoparticles embedded in such films were used as catalyst for the hydrogenation of nitroarenes. The catalytic performance test shows that the thin film catalyst can be applied to the hydrogenation reaction of various substituted nitroaromatics, and exhibits good catalytic activity and good catalyst stability. It is worth mentioning that our catalytic films can be easily removed from the reaction system in any time during the reaction, and the catalytic activity could be fully recovered when reused directly in another catalytic cycle for five times.

Metal-directed self-assembly has been applied to prepare supramolecular coordination polygons which adopt tetrahedral (1) or trigonal disklike topologies (2). In the solid state, 2 assembles into a stable halide-metal-organic material (Hal-MOM-2), which catalyzes H2O oxidation under photo- and electrocatalytic conditions, operating with a maximum TON = 78 and TOF = 1.26 s(-1). A facile and general synthesis of 2,4,6-triaryl pyrimidines has been developed. It involves a one-pot [2+1+1+1+1] pseudo five-component annulation of one methyl ketone, two aldehydes and two NH4OAc catalyzed by TfOH. One C-C and four C-N bonds are formed during the oxidative annulation process. The reaction shows good tolerance of many important functional groups in air and produces only water as the coproduct, making this methodology a highly versatile alternative to the existing methods for structuring pyrimidine framework.

We have developed an iron-catalyzed cyclization reaction between ketoxime carboxylates and nitrones to afford trisubstituted pyridines.
