Chemoselective hydrogenation of nitroarenes to anilines is of great importance for the manufacture of pharmaceuticals, fine chemicals, and dyes. In this study, a series of metal-free N-doped carbon nanotubes (NCNTs) have been prepared by the carbonization of in situ polymerized pyrrole on CNTs. The concentration of pyrrole, pyrolysis temperature, and the outside diameter of CNTs were investigated to improve the catalytic performance. As characterized by Raman and X-ray photoelectron spectroscopy (XPS), the optimal catalyst (NCNTs-800) possessed a unique structure doped with the same content of pyrrolic N and graphitic N. The activity and selectivity of NCNTs-800 have been evaluated for the selective hydrogenation of substituted nitroarenes. Highly selective hydrogenation of the nitro group of 12 different substrates has been achieved on NCNTs-800, even in the presence of a fragile iodo group. The hydrogenation reaction on N-doped CNTs from polypyrrole involved a mixture of different hydrogen species including nonpolar H radicals. In addition, stability and recyclability of NCNTs-800 have been tested.

The regioselective N-addition/substitution reaction between alpha-alkylidene pyrazolinones and propargyl sulfonium salts has been developed to construct functionalized allylthio-containing pyrazolones with moderate to excellent yields. alpha-Alkylidene pyrazolinones act as N-nucleophilic agents which are distinguished from reported C-nucleophilic reactions. The secondary amine participating asymmetric reductive amination remains an unsolved problem in organic synthesis. Here we show for the first time that secondary amines are capable of effectively serving as N-sources in direct asymmetric reductive amination to afford corresponding tertiary chiral amines with the help of a selected additive set under mild conditions (0-25 degrees C). The applied chiral phosphoramidite ligands are readily prepared from BINOL and easily modified. Compared with common tertiary chiral amine synthetic methods, this procedure is much more concise and scalable, as exemplified by the facile synthesis of rivastigmine and N-methyl-1-phenylethanamine.

Zr-based metal-organic frameworks have been considered promising heterogeneous catalysts in various reactions. It has been proven that the defect sites in these frameworks, which result from deficiency of linkers, could act as the reaction active sites. In this work, microwave irradiation was used for rapid synthesis and increasing defects on Zr-6 clusters of metal-organic framework UiO-66. Powder X-ray diffraction, thermal gravimetric analysis, and N-2 sorption isotherms were performed to characterize and calculate the defect number in the framework. UiO-66 prepared by microwave method for a few minutes could reach 1.8 missing linkers per cluster. The material with acetic acid as a co-catalyst showed highly heterogeneous catalytic activity in the solvent-free heterocyclization of 2,2-disubstituted 2,3-dihydroquinazolin-4(1H)-ones from anthranilamide and ketones/aldehydes. The synthetic scheme consisted of zirconium cluster catalysed carbonyl group of anthranilamide to achieve the C-N propitious combination and succeeding ring closure process.

In the present study, we report the design and synthesis of novel CAN508 sulfonamide-based analogues (4, 8a-e, 9a-h and 10a-e) as novel carbonic anhydrase (CA) inhibitors with potential CDK inhibitory activity. A bioisosteric replacement approach was adopted to replace the phenolic OH of CAN508 with a sulfamoyl group to afford compound 4. Thereafter, a ring-fusion approach was utilized to furnish the 5/5 fused imidazopyrazoles 8a-e which were subsequently expanded to 6/5 pyrazolopyrimidines 9a-h and 10a-e. All the synthesized analogues were evaluated for their inhibitory activity toward isoforms hCA I, II, IX and XII. The target tumor-associated isoforms hCA IX and XII were effectively inhibited with KIs ranges 6-67.6 and 10.1-88.6 nM, respectively. Furthermore, all compounds were evaluated for their potential CDK2 and 9 inhibitory activities. Pyrazolopyrimidines 9d, 9e and 10b displayed weak CDK2 inhibitory activity (IC50 = 6.4, 8.0 and 11.6 mu M, respectively), along with abolished CDK9 inhibitory activity. This trend suggested that pyrazolopyrimidine derivatives merit further optimization to furnish more effective CDK2 inhibitor lead. On account of their excellent activity and selectivity towards hCA IX and XII, pyrazolopyrimidines 10 were evaluated for their anti-proliferative activity toward breast cancer MCF-7 and MDA-MB-468 cell lines under normoxic and hypoxic conditions. The most potent anti-proliferative agents 10a, 10c and 10d significantly increased cell percentage at sub-G1 and G2-M phases with concomitant decrease in the S phase population in MCF-7 treated cells. Finally, a docking study was undertaken to investigate the binding mode for the most selective hCA IX and XII inhibitors 10a-e, within hCA II, IX and XII active sites. (C) 2020 Elsevier Masson SAS. A 4-HO-TEMPO-catalyzed redox strategy for the synthesis of pyridines through the annulation of cyclopropanols and oxime acetates has been developed. This protocol features good functional group tolerance and high chemoselectivity and also promises to be efficient for the late-stage functionalization of skeletons of drugs and natural products. Mechanism studies indicate that the reaction involves the in situ generated alpha,beta-unsaturated ketones and imines as the key intermediates, which are derived from cyclopropanols and oxime acetates via a TEMPO/TEMPOH redox cycle, respectively. The pyridine products are formed as a result of annulation of enones with imines followed by TEMPO-catalyzed oxidative aromatization by excess oxime acetates. This method not only realizes the TEMPO-catalyzed redox reaction but also broadens the frontiers for TEMPO in catalysis.

Summary of main observation and conclusion Selenium and copper synergistically catalyzed the oxidation/alkoxylation of methyl ketones to synthesize alpha-keto acetals directly. Using O(2)as oxidant and alcohol as solvent and alkoxylation reagent, the reaction is practical from industrial viewpoint. Graphitic carbon nitride (g-C3N4, CN for short) is a compelling visible-light responsive photocatalyst. However, its photocatalytic efficiency is low due to the random carrier transfer in planes and insufficient redox potential. Herein, we build oxygen functional group modified sodium-doped carbon nitride (OH/Na co-functionalized carbon nitride) to promote directional transfer of charge carriers for acceleration of separation and enhance redox potential for efficient oxidation of NO in air. Specifically, the function of sodium atoms could control the directional transfer of random carriers from the intralayer to the oxygen functional group-modified surface for the purpose of effectively reducing photogenerated electron-hole recombination. Meanwhile, the modification by oxygen-containing functional groups could adjust the band structure of CN, thereby increasing the oxidation-reduction potential of NO in the photocatalyst. The transformation pathways and reaction mechanism of photocatalytic NO oxidation on CN and OH/Na co-functionalized carbon nitride have also been explicated by ESR spectroscopy and in situ DRIFTS and compared. This work provides a new method for simultaneously controlling the random transfer of carriers and adjusting the energy band structure of CN to optimize its photocatalytic efficiency. It is also possible to extend this strategy to improve the performance of other 2D layered catalysts for photocatalytic oxidation.

In synthetic chemistry, the protection of aldehydes and ketones is crucial during multistep synthesis of complex molecules. Organic chemists have paid substantial attention to the synthesis of 1,3-oxathiolanes and 1,3-oxathianes because of their considerable stability under acidic conditions and ease of removal of protecting groups. In this paper, we report the mild and efficient oxathiacetalization of aldehydes with 2-mercaptoethanol and 3-mercaptopropan-1-ol through visible-light-promoted eosin-Y catalyzed C-S and C-O bond formation at ambient temperature under metal-free conditions. This catalytic system also affords oxathiacetalization of ketones through photoredox catalysis.
