A strategy for heterogeneous copper(I) mesoporous silica [Cu(I)-SBA-15] mediated catalytic reduction has been developed for the reduction of various nitroarenes in excellent yields. In addition to alcothermal strategy, other strategies have been attempted for the preparation of Cu(I) substituted mesoporous silica, however, the desired oxidation state is obtained only through alcothermal strategy. The crystalline phases and oxidation state of synthesized materials were characterized by XRD and XPS measurements and tested for catalytic reduction of nitroarenes. Among the tested materials, the best catalytic reduction of nitroarenes with an excellent substrate scope and good catalytic reusability was established by Cu(I)-SBA-15 material. The reduction kinetics and the controlled experimental studies reveal that the reaction proceeds through pseudo-first-order kinetics and nitrosobenzene intermediate pathway.

We report an iodoarene-catalyzed enantioselective synthesis of beta,beta-difluoroalkyl bromide building blocks. The transformation involves an oxidative rearrangement of alpha-bromostyrenes, utilizing HF-pyridine as the fluoride source and m-CPBA as the stoichiometric oxidant. A catalyst decomposition pathway was identified, which, in tandem with catalyst structure-activity relationship studies, facilitated the development of an improved catalyst providing higher enantioselectivity with lower catalyst loadings. The versatility of the difluoroalkyl bromide products was demonstrated via highly enantiospecific substitution reactions with suitably reactive nucleophiles. The origins of enantioselectivity were investigated using computed interaction energies of simplified catalyst and substrate structures, providing evidence for both CH-pi and pi-pi transition state interactions as critical features.

Selective monoamine oxidase B inhibitors are potential drug candidates for the treatment of Parkinson’s disease. A series of phenyl substituted isoxazole carbohydrazides was designed by structural modification of isocarboxazid, a nonselective MAO inhibitor and evaluated as inhibitors of MAO-A and MAO-B. The compounds were not able to inhibit MAO-A significantly, but most of the compounds exhibited potent inhibitory activity against MAO-B. The enzyme kinetic study of the most active compounds 5d, 5-phenyl-N ‘-(1-(p-tolyl)ethylidene)isoxazole-3-carbohydrazide and 5g, N ‘-(1-(3,4-dimethoxyphenyl)ethylidene)-5-phenylisoxazole-3-carbohydrazide displayed reversible and competitive MAO-B inhibition. In molecular modeling studies, compounds 5d and 5g exhibited strong binding affinity on MAO-B active site. The administration of compounds 5d and 5g exhibited prevention of MPTP-induced Parkinsonism as indicated by footprint analysis and horizontal wire tests. Further optimization studies are essential to exploit their potential for MAO-B associated neurodegenerative pathologies.

In the current study, a series of macrocyclic tripeptide Schiff bases 2-6 were synthesized by using macrocyclic tripeptide hydrazide 1 as starting material. The selective hydrogenation of nitro compounds into their corresponding azo compounds is challenging in organic synthesis. In this study, we have developed a facile and efficient catalytic system for the transformation of nitro compounds into azo compounds under mild conditions over a heterogeneous cobalt catalyst (Co-N-x/C-800-AT). The Co-N-x/C-800-AT catalyst was found to be highly active for the reductive-coupling of nitro compounds into azo compounds in ethanol at 110 degrees C and 2.5 bar H-2 pressure with the addition of NaOH as an additive, affording structurally-diverse azo compounds with satisfactory yields (70.4-98.3%). The developed method demonstrated several advantages such as high catalytic activity and high selectivity with tolerance to other functional groups and high stability.
