Development of new highly potent imidazo[1,2-b[pyridazines targeting Toxoplasma gondii calcium-dependent protein kinase 1

Using a structure-based design approach, we have developed a new series of imidazo[1,2-b]pyridazines, targeting the calcium-dependent protein kinase-1 (CDPK1) from Toxoplasma gondii. Twenty derivatives were thus synthesized. Structure-activity relationships and docking studies confirmed the binding mode of these inhibitors within the ATP binding pocket of TgCDPK1. Two lead compounds (16a and 16f) were then identified, which were able to block TgCDPK1 enzymatic activity at low nanomolar concentrations, with a good selectivity profile against a panel of mammalian kinases. The potential of these inhibitors was confirmed in vitro on T. gondii growth, with EC50 values of 100 nM and 70 nM, respectively. These best candidates also displayed low toxicity to mammalian cells and were selected for further in vivo investigations on murine model of acute toxoplasmosis.

Synthesis and urease inhibitory activity of 1,4-benzodioxane-based thiosemicarbazones: Biochemical and computational approach

The hyperactivity of urease enzyme is associated with clinically important complications including stomach ulcers and kidney stones. This enzyme provides a suitable environment to Helicobacter pylori at low pH in the stomach, a causative agent of peptic and ulcer gastric that may lead to cancer. Natural and synthetic small molecules were reported to inhibit urease enzyme. Within this context, a new series of 1,4-benzodioxane-based thiosemicarbazones (3a-p) were synthesized and screened in vitro against urease enzyme to elucidate their anti-urease activity. All the compounds displayed potent inhibitory potential with IC50 values ranging between 3.65 ± 2.64 to 31.9 ± 1.094 muM, under positive control of thiourea (IC50 = 20.8 ± 0.75 muM). Structural activity relationship (SAR) has revealed a variation based on substituents pattern at R group. The results of docking study suggest that these compounds thermodynamically binds via nickel atoms present in the active-site of urease. The in silico docking analysis and our experimental findings are in excellent co-relation. The pharmacokinetic behavior of all the compounds were also predicted.

The extraction residue from Shengli lignite was sequentially dissolved with cyclohexane, benzene, methanol, ethanol, and isopropanol in an autoclave at 320 C to afford soluble portions (SPs) 1?5 (SP1-SP5) and the final residue (FR). The total yield of SP1-SP5 is ca. 55.1%. FR was subjected to ruthenium ion-catalyzed oxidation and the resulting products were isolated from the reaction mixture and esterified. Both the esterified products and SP1-SP5 were analyzed with a gas chromatograph/mass spectrometer. In total, 342 compounds were identified in SP1-SP5. They can be classified into normal alkanes, branched alkanes, alkenes, alkanedienes, arenes, alkanols, methylcycloalkanes, alkenols, alkylbenzenemethanols, arenols, anisol and substituted anisols, polymethyldihydrobenzofurans, arenofurans, dibenzofurans, ethoxymethylbenzenes, aldehydes, ketones, esters, nitrogen-containing organic compounds, sulfur-containing organic compounds, and other compounds. Among the compounds, arenols are predominant in SP1 and SP2 and the main compounds in SP3, while the main compounds in SP4 and SP5 are esters and arenes, respectively. According to the esterified products identified, the products from FR oxidation can be grouped into non-benzene ring carboxylic acids (NBCAs) and benzenepolycarboxylic acids (BPCAs). The total yield of BPCAs is much higher than that of NBCAs, suggesting that FR is rich in condensed aromatic moieties.

Enaminone-Derived Pyrazoles with Antimicrobial Activity

A series of pyrazoles derived from the substituted enaminones were synthesized and were evaluated for antimicrobial activity. All the compounds were characterized by the spectral data and elemental analysis. The synthesized compounds were initially screened for their antimicrobial activity against ATCC 6538, NCTC 10400, NCTC 10418, and ATCC 27853. During initial screening, compounds (P1, P6, and P11) presented significant antimicrobial activity through disc diffusion assay. These compounds were further evaluated for antimicrobial activity at different time points against Gram-positive and Gram-negative bacteria and presented significant activity for 6 hours. The activity was found to be greater against Gram-positive bacteria. In contrast at 24 hours, the activity was found only against Gram-positive bacteria except compound (P11), showing activity against both types of bacteria. Compound (P11) was found to have highest activity against both Gram-positive and Gram-negative bacteria.
