On the pharmaceutical account of the reported anticancer activity of benzothiazole derivatives, differently substituted benzothiazole derivatives 2a-c to 34a,b, attached at 2-position to different heterocyclic moieties, were synthesized via different chemical reactions. Thirteen of the newly synthesized compounds were selected by the National Cancer Institute, Bethesda, Maryland, USA, and evaluated for their in vitro antitumor activity against 60 human tumor cell lines in a one-dose screening panel among which two compounds 4 and 17 showed high activity and were selected for further evaluation in the five-dose full panel assay, in which compound 4 exerted powerful growth inhibitory activity against all cell lines with GI(50) ranging from 0.683 to 4.66 mu M/L in addition to excellent lethal activity against most of the cell lines.

A copper-catalyzed approach for the N-acylation of anilines with acetone and acetophenones via C-C bond cleavage is described. Under the developed conditions both CHCl3 and CH2Cl2 were identified as potential C1-source to promote the transformation. The reaction features a site selective C-C bond cleavage to install the amide moieties with high functional-group compatibility and wide substrate scope. The developed method avoids the use of sensitive and narcotic agents. The method also represents an excellent complement to the previous protocols with lower E-factor (13.91 mg/1 mg) than current industrially used method (E-factor 17.54 mg/1 mg). The developed approach has also been extended for the effective preparation of pyridine derivatives and paracetamol in gram scale. The course of the reaction was monitored by H-1 NMR as a preliminary investigation of the reaction mechanism.

We have prepared 15 hybrid pyrazole, pyrazoline-clubbed pyridine-containing compounds (5a-o) and tested for their antibacterial and antifungal activities for the development of potential antimicrobial agents. The structures of this novel series were characterized by various spectral techniques like IR, H-1 NMR, C-13 NMR, LC-MS, and elemental analysis. The synthesized compounds 5d, 5e, 5i, 5k, 5m, and 5o exhibited significant antimicrobial activity in the comparison of standard drugs. Molecular docking studies that have been carried out to emphasize the binding orientations of these molecules were in good compliance with crystal structure interactions. The predicted drug-likeness (ADME) properties were found to be in the acceptable range.

A practical method for the oxidation of aromatic side chains was established for the preparation of aromatic aldehydes and ketones. Using NiCl2 as the catalyst, substituted toluenes were oxidized with Na2S2O8 at the benzvlic site for the synthesis of the corresponding aldehydes in the yield of 22%similar to 79%. Ethylbenzene analogs were oxidized more easily to obtain the corresponding ketones with 64%similar to 84% yields. The oxidation of benzyl alcohol analogs was completed to acquire the corresponding carbonyl compounds in shorter time with better selectivity and yields. The method has the advantages such as the mild reaction conditions, no requirement for precious metals or additional promoter, and good selectivity.

Hydrogen peroxide (H2O2) is a small molecule of reactive oxygen species, mainly produced by cell mitochondria, which plays an important role in physiological processes. Excess of H2O2 in normal cells could causes a variety of diseases. For that reason, it is very significant to emerge a detection method with high-precision for H2O2. Herein, we reported a structurally novel fluorescent probe, para-nitrophenyl oxoacetyl rhodamine (RhB-NIR) for the detection of H2O2 levels in living cells. The results show that the Stokes shift for RhB-NIR is 140 nm, much larger than those for other reported rodamine derivatives. RhB-NIR shows satisfactory specificity and high sensitivity for the detection of H2O2. Excellent linear relationship with H2O2 within a certain concentration range (0-10 mu M) was displayed, and the limit of detection (LOD) is 61 nM. More importantly, RhB-NIR realized fluorescence imaging of H2O2 in living cells. In addition, RhB-NIR also shows great potential for targeting mitochondria in cell colocalization experiments, in which the Pearson’s coefficient is 0.91 using Rhodamine 123 as a reference. This probe will be a promising tool for detecting H2O2 in the future research.

We have developed an efficient metal-free route for the synthesis of 1,4-enedione derivatives under microwave irradiation by reacting easily available arylmethylketones with DMSO or diphenyl sulfoxide in the presence of TBAI and persulfate. The highly diastereoselective synthesis of perfluoroalkyl-containing pyrano[3,4-c]pyrroles has been accomplished via a cascade process involving Michael addition, Passerini-type reaction, Mumm rearrangement and an oxo-Diels-Alder reaction. A new, cheap, and accessible method has been used for the preparation of nano-belts from the chemical decomposition (top-down approach) of a cheap stainless steel screw and found as an efficient magnetically recyclable nanocatalyst for the preparation of quinolines and 1,8-dioxo-octahydroxanthenes under mild reaction conditions. The nano-belts, Fe-Cr-Ni oxide alloy, was prepared in a two-step synthesis and characterized with various instrumental methods. Due to magnetic property of the screw (a ferritic-alloy), the resultant nano-belts is magnetic. Magnetic Fe-Cr-Ni alloy nano-belts were applied toward efficient preparation of quinolines and 1,8-dioxo-octahydroxanthenes under mild conditions. The catalyst could be readily recovered and recycled for several consecutive runs, while it suffers from a very low metal leaching and subsequently efficiency drop. Graphic abstract
