Cross-dehydrogenative C-O coupling of N-hydroxyimides with ketones, esters, and carboxylic acids was achieved employing the di-tertbutyl peroxide as a source of free radicals and a dehydrogenating agent. The proposed method is experimentally simple and demonstrates the outstanding efficiency for the challenging CH substrates, such as unactivated esters and carboxylic acids. It was shown that N-hydroxyphthalimide drastically affects the oxidative properties of t-BuOOt-Bu by intercepting the t-BuO center dot radicals with the formation of phthalimide-N-oxyl radicals, a species responsible for both hydrogen atom abstraction from the CH reagent and the selective formation of the C-O coupling product by selective radical cross-recombination. The practical applicability of the developed method was exemplified by the single-stage synthesis of commercial reagent (known as Baran aminating reagent precursor) from isobutyric acid and N-hydroxysuccinimide, whereas in the standard synthetic approach, four stages are necessary.

A copper-catalyzed approach for the N-acylation of anilines with acetone and acetophenones via C-C bond cleavage is described. Under the developed conditions both CHCl3 and CH2Cl2 were identified as potential C1-source to promote the transformation. The reaction features a site selective C-C bond cleavage to install the amide moieties with high functional-group compatibility and wide substrate scope. The developed method avoids the use of sensitive and narcotic agents. The method also represents an excellent complement to the previous protocols with lower E-factor (13.91 mg/1 mg) than current industrially used method (E-factor 17.54 mg/1 mg). The developed approach has also been extended for the effective preparation of pyridine derivatives and paracetamol in gram scale. The course of the reaction was monitored by H-1 NMR as a preliminary investigation of the reaction mechanism.

Integrated continuous manufacturing has emerged as a promising device for the rapid manufacturing of active pharmaceutical ingredients (APIs). We herein report a newly designed continuous flow micro-total process system platform for the rapid manufacturing of celecoxib, a selective nonsteroidal anti-inflammatory drug. This approach has been proven generally for the synthesis of several alkyl and aryl substituted pyrazoles. In order to minimize the tedious work-up process of potential reaction intermediates/products, we have developed a continuous flow extraction and separation platform to carry out the entire reaction sequence resulting in a short residence time with good yield. The present process was further extended to gram scale synthesis of the COX-2-related API, viz. celecoxib, in the continuous flow process.

Herein, we demonstrate an efficient protocol for transfer hydrogenation of ketones using methanol as practical and useful liquid organic hydrogen carrier (LOHC) under Ir(III) catalysis. The crystal structures of seven alpha-aryl-alpha-hydroxyphosphonates synthesized by the Pudovik reaction of substituted benzaldehydes and dialkyl phosphites, namely dimethyl [(hydroxy)(phenyl)methyl] phosphonate, C9H13O4P, dimethyl [(3,4-dimethoxyphenyl)(hydroxy)methyl]phosphonate, C11H17O6P, dimethyl (1-hydroxy-1-phenylethyl) phosphonate, C10H15O4P, dimethyl [1-hydroxy-1-(4-nitrophenyl)ethyl]phosphonate, C10H14NO6P, dibenzyl [hydroxy(2-nitrophenyl)methyl]phosphonate, C21H20NO6P, dibenzyl [(3-chlorophenyl)(hydroxy)methyl]phosphonate, C21H20ClO4P, and dibenzyl [hydroxy(4-methylphenyl)methyl]phosphonate, C22H23O4P, were studied to gain a better understanding of the organization in this type of molecule in the solid state. The crystals obtained for this series of compounds show a balance between C-OH center dot center dot center dot O-P chain-linked packing and the dimeric types of hydrogen-bond bridges of intermolecular pairs of such functions. The description is based on primary graph-set descriptors. Using graph-set descriptors one level deeper (i.e. secondary graph sets of the C-H center dot center dot center dot O type) revealed a similarity in the graph-set descriptors, suggesting a fine interplay of substituent- and shape-dependent effects on strong-weak interactions. It seems that the formation of chains or dimers is governed not only by the presence of a tertiary C alpha atom, but also by the nature and crowding of the ortho substituents of the alpha-aryl group.

Nitro-reduction of aromatic nitro compounds into anilines has vast opportunities in synthetic chemistry. The catalytic systems used earlier operated under harsh conditions, which was very expensive to achieve end products. Visible light has advantages over the catalytic system due to the easy availability, low cost, and abundance. The present methodology describes the efficient reduction of a variety of aromatic nitro compounds into the corresponding anilines using a Z-scheme Fe2O3/BiVO4 photocatalyst under visible light illuminance. The photocatalyst demonstrated excellent activity up to 99% conversion of the nitrobenzene into aniline and for a series of nitroarenes as well at room temperature and atmospheric pressure visible light irradiation. The catalytic system has the advantage due to no leaching in the recycling experiments and high stability up to six cycles. The use of low-cost transition and p-block metal enhances the applicability of the catalytic system for aromatic nitro compounds reduction into the corresponding amines.

Modification of active metals with metal oxide modifiers has attracted considerable attention in heterogeneous catalysis due to their synergistic effect. However, a controllable synthesis of highly reactive and stable metal-metal oxide hybrid nanocatalysts is difficult. To solve this problem, presynthesized IrM (M = Fe, Co, and Ni) bimetallic nanoparticles were initially confined in the mesopores of SBA-15 and were then in situ transformed to Ir-MOx hybrids. The obtained Ir-MOx/SBA-15 nanocatalysts show superior activity and selectivity in the hydrogenation of substituted nitroaromatics to corresponding aromatic amines compared to Ir/SBA-15. Among these Ir-based catalysts, Ir-FeOx/SBA-15 exhibits the highest activity and selectivity and has a wider substrate scope due to the interaction between Ir and FeOx. Therefore, our research provides a way to design reactive and stable hydrogenation catalysts.
