Synthesis of a series of 2-arylbenzo[d]imidazo[2,1-b] thiazoles tethered with barbituric acid moiety has been reported from the three component reaction of 2-aminobenzothiazoles, barbituric acids and terminal aryl acetylenes or aryl methyl ketones in the presence of I-2 in DMSO medium. Both conventional and microwave heating conditions can be used for this multicomponent reaction. The salient features of this methodology are: (i) formation of one C-C and two C-N bonds in one-pot under metal-free oxidation followed by cyclization, (ii) selective formation of the fused imidazole ring, (iii) wide substrate scope, (iv) easy purification of the products, (v) products having more than one pharmaceutically important motifs and (vi) gram scale synthesis possible.

Substituted phenylthiazolyl benzamide and pyrrolyl benzamide derivatives were developed using molecular hybridization technique to create novel lead antimycobacterial molecules used to fight against Mycobacteriumtuberculosis. The newly synthesized molecules have inhibited InhA, the enoyl-ACP reductase enzyme from the mycobacterial type II fatty acid biosynthetic pathway. Of these, compound 3b showed H-bonding interactions with Tyr158 and co-factor NAD(+) that binds the active site of InhA. All the molecules were screened for in vitro antitubercular activity against M. tuberculosis H(37)Rv, as well as some representative molecules as the inhibitors of InhA. Thirteen compounds exhibited good anti-TB activities (MIC = 1.6 mu g/mL), but only few representative molecules showed the moderate InhA enzyme inhibition activity. Basic core moiety and docked mode of all the synthesized compounds inside the proposed binding pocket of InhA with the final selected pharmacophore model molecular alignment for InhA receptor ligands. A copper-mediated domino condensation reaction of readily accessible oxime acetates with ninhydrin is reported to afford pyrrolo[2,1-a]isoindolediones via new C-C & C-N bond formations. A wide range of oxime acetates were shown to generally participate in the reaction to produce the condensed products in excellent yields. The necessary control experiments were performed and the mechanism is proposed to involve sequentially the formation of iminium radical via Cu-mediated N-O bond cleavage of oxime acetates, addition of the radical to ninhydrin and rearrangement via ring expansion.

Thirty-eight chalcone derivatives bearing a chromen or benzo[f]chromen moiety were synthesized and evaluated for their anti-inflammatory and analgesic activities. Using an ear edema model, anti-inflammatory activities were observed for compounds 3a-3s (ear inflammation: 1.75-3.71 mg) and 4a-4s (ear inflammation: 1.71-4.94 mg). All compounds also displayed analgesic effects with inhibition values of 66.7-100% (3a-3s) and 96.2-100% (4a-4s). The 12 compounds that displayed excellent anti-inflammatory and analgesic effects were tested for their inhibitory activity against ovine COX-1 and COX-2. Six compounds bearing a chromen moiety were weak inhibitors of the COX-1 isozyme but showed moderate COX-2 isozyme inhibitory effects (IC(50)s from 0.37 mu M to 0.83 mu M) and COX-2 selectivity indexes (SI: 22.49-9.34). Those bearing a benzo[f]chromen moiety were more selective toward COX-2 than those bearing a chromen moiety with IC(50)s from 0.25 mu M to 0.43 mu M and COX-2 selectivity indexes from SI: 31.08 to 20.67.

Sustainable development demands an environmentally friendly and efficient method for the hydrogenation of organic molecules, including the hydrogenation of functionalized nitroarenes. In this study, a highly active and selective metal-organic framework-supported palladium catalyst was prepared for the catalytic hydrogenation of nitroarenes. High selectivity (>99%) and excellent yield (98%) of aniline were realized after 2 hours in ethanol under hydrogen (1 atm) at room temperature. The reductions were successfully carried out in the presence of a wide range of other reducible functional groups. More importantly, the catalyst was very stable without the loss of its catalytic activity after five cycles.

Hydrofunctionalization, the direct addition of an X-H (e.g., X=O, N) bond across an alkene, is a desirable strategy to make heterocycles that are important structural components of naturally occurring molecules. Described here is the design and discovery of donor-acceptor-type platinum catalysts that are highly effective in both hydroalkoxylation and hydroamination of unactivated alkenes over a broad range of substrates under mild conditions. A number of alkene substitution patterns are accommodated, including tri-substituted, 1,1-disubstituted, (E)-disubstituted, (Z)-disubstituted and even mono-substituted double bonds. Detailed mechanistic investigations suggest a plausible pathway that includes an unexpected dissociation/re-association of the electron-deficient ligand to form an alkene-bound donor-acceptor-type intermediate. These mechanistic studies help understand the origins of the high reactivity exhibited by the catalytic system, and provide a foundation for the rational design of chiral catalysts towards asymmetric hydrofunctionalization reactions. The direct addition of an O-H (hydroalkoxylation) or an N-H (hydroamination) bond across an alkene is a useful strategy to access biologically relevant heterocycles. Here, the authors report donor-acceptor-type platinum catalysts that are effective for both hydroalkoxylation and hydroamination of unactivated alkenes, with broad substrate scope and use of mild conditions.

Modified polycyanostyrene and polynitrostyrene microspheres were designed and synthesized, which proved to be an effective carrier to prepare gold nanoparticles with pentazole gold as a catalyst precursor. The developed gold nanoparticles were characterized using scanning electron microscopy, transmission electron microscopy, energy dispersive X-ray, X-ray power diffraction and X-ray photoelectron spectrometry. In addition, it was observed that this catalytic system successfully achieved application in the synthesis of cinnoline derivatives and hydration reactions in high yields. Mechanism investigation was conducted to better understand these transformations and catalyst systems. To the best of our knowledge, this is the first example of utilizing pentazole gold as the catalyst precursor and achieving catalytic applications.

In the present investigation, an alluring green PEG-400 mediated one-pot synthesis of novel 2-(2-hydrazinyl)thiazole derivatives from 1,2,6,7-tetrahydro-8H-indeno[5,4-b]furan-8-one has been unveiled via a novel synthetic pathway. The application of PEG in organic reactions as a reaction media is one of the fantastic tools of green chemistry as reactions can be carried out under generous conditions limiting environmental peril and chemical waste. PEG-400 is recognized as a low-cost, non-flammable, environmentally benign, recyclable, and richly available green solvent. A series of novel 2-(2-hydrazineyl)thiazole derivatives have been synthesized in good to excellent yield by using the green capability of PEG-400 solvent. These newly synthesized compounds were tested for their antimicrobial and antioxidant activities. The results revealed that these compounds show good activities compared with the standard. Additionally, all the synthesized compounds exhibit negligible cytotoxicity as compared to the positive control. The structures of all novel compounds reported herein are established using FT-IR, H-1 NMR, C-13 NMR, D2O exchange experiment and HRMS techniques.
