A convenient one-pot, three-component, and solvent-free procedure for the preparation of substituted (E)-1-phenyl-3-[2-(piperidin-1-yl)quinolin-3-yl]prop-2-en-1-ones, which has made a significant improvement of previously reported methods, has been developed. The reaction of chloro aldehydes, ketones, and piperidine under microwave irradiation afforded the corresponding piperidine-substituted chalcone derivatives in high yields in shorter reaction times. All the synthesized compounds were characterized by IR and H-1 and C-13 NMR spectroscopy, and mass spectroscopy.

A series of novel chalcone derivatives pyrido[4,3-b]pyrazin-5(6H)-one (10a-j) were designed, synthesized and their structures were confirmed by H-1 NMR, C-13 NMR and mass spectral data. Further, all derivatives were tested for their anticancer activities against five human cancer cell lines such as MCF-7 (breast cancer), A-549 (lung cancer), Colo-205 (colon cancer), A2780 (ovarian cancer) and DU-145 (prostate cancer) by employing MIT assay. The clinically used drug etoposide was used as standard reference and the anticancer activity was expressed as the IC50 in mu M. Among the ten compounds examined compounds, 10b, 10c, 10d, 10h, and 10i possessed more promising anticancer activity. A molecular docking study implying ATR kinase was carried out to observe the binding mode of chalcone derivatives pyrido[4,3-b]pyrazin-5(6H)-one (10a-j) on the active site of ATR kinase. The most promising compound, 10h showed pi – pi stacking interaction of trimethoxy phenyl and pyridone moiety with the residue Trp850, while the carbonyl (alpha,beta-unsaturated carbonyl) and methoxy functions showed H-bond interaction with the residue Ser773 and Thr856 respectively. Chemical diversification of type II topoisomerase (Topo II) inhibitors remains indispensable to extend their anti-tumor therapeutic values which are limited by their side effects. Herein, we designed and synthesized a novel series of benzimidazole-chalcone hybrids (BCHs). These BCHs showed good inhibitory effect in the Topo II mediated DNA relaxation assay and anti-proliferative effect in 4 tumor cell lines.4dand4nwere the most potent, with IC(50)values less than 5 mu M, superior to etoposide. Mechanistic studies indicated that the BCHs functioned as non-intercalative Topo II catalytic inhibitors. Moreover,4dand4ndemonstrated versatile properties against tumors, including inhibition on the colony formation and cell migration, and promotion of apoptosis of A549 cells. The structure-activity relationship and molecular docking analysis suggested possible contribution of the chalcone motif to the Topo II inhibitory and anti-proliferative potency. These results indicated that4dand4ncould be promising lead compounds for further anti-tumor drug research.

In the present study, an innovative cold plasma treatment was used as a tool to fortify the white rice. The amounts of ferrous sulphate and ascorbic acid were optimized for improving the bioavailability of iron. White rice samples were treated with plasma at a constant voltage of 20 kV for varying time (10 min and 15 min). The exposure time of plasma was selected based on the surface characteristics, hydrophilicity and thermal properties. Significant improvement was observed in the characteristics of hydrophilicity, surface energy, cooking time and hardness of plasma-treated rice. Plasma treated rice was fortified with the optimum concentrations of iron and ascorbic acid solution. Optimum concentrations of iron and ascorbic acid per 100 g of rice (862.93 mg and 1398.27 mg) were found by conducing experiments using Central Composite Design of Response Surface Methodology. Further, rice was blended with untreated rice in the ratio of 1:100 and 1:200 and was packed in LDPE and PP pouches and was stored at ambient temperature for further storage analysis. The in vitro bioavailability of iron was significantly higher in the plasma-treated fortified rice at both 1.35 and 7.5 pH than in the control sample, and plasma treatment significantly reduced the rate of oxidation of iron during storage.

Selective transfer hydrogenation of nitroarenes to amines with transition metal nanocatalysts is appealing due to its low-cost, moderate reaction conditions, good activity and excellent selectivity. Single-atom catalysts (SACs) possessing advantages of maximum atom efficiency and particular electronic structure are expected to be more effective for this reaction, yet no report about it. Herein, cobalt single atoms anchored on N-doped ultrathin carbon nanosheets (denoted as CoSAs/NCNS) were produced and demonstrated as an outstanding SAC for selective transfer hydrogenation of nitroarenes to amines with formic acid as hydrogen donor. The turnover frequency (TOF) reached 110.6 h(-1), which was 20 times higher than the best results of cobalt nanoparticles reported in literatures under similar reaction conditions. Moreover, CoSAs/NCNS exhibited excellent selectivity for a variety of nitroarenes bearing other reducible functionalities, such as iodo, cyano, keto, vinyl, alkynyl and ester groups. The findings further highlight the ability and advantages of SACs in heterogeneous catalysis.

In this study, a series of 20 chalcone derivatives was synthesized, and their antiproliferative activity was tested against the human T cell acute lymphoblastic leukemia-derived cell line, CCRF-CEM. On the basis of the structural features of the most active compounds, a new library of chalcone derivatives, according to the structure-activity relationship design, was synthesized, and their antiproliferative activity was tested against the same cancer cell line. Furthermore, four of these derivatives (compounds 3, 4, 8, 28), based on lower IC50 values (between 6.1 and 8.9 mu M), were selected for further investigation regarding the modulation of the protein expression of RACK1 (receptor for activated C kinase), protein kinase C (PKC)alpha and PKC beta, and their action on the cell cycle level. The cell cycle analysis indicated a block in the G0/G1 phase for all four compounds, with a statistically significant decrease in the percentage of cells in the S phase, with no indication of apoptosis (sub-G0/G1 phase). Compounds 4 and 8 showed a statistically significant reduction in the expression of PKC alpha and an increase in PKC beta, which together with the demonstration of an antiproliferative role of PKC beta, as assessed by treating cells with a selective PKC beta activator, indicated that the observed antiproliferative effect is likely to be mediated through PKC beta induction.

y A novel polymeric network benefiting from the chemistry of imidazolium salt (IL), cyclodextrin (CD) and carbon nanotube (CNT) is fabricated through a multi-step process, in which silica coated CNTs were vinyl functionalized and polymerized with poly (ethylene glycol) dimethacrylate and CD-IL, prepared from the reaction of vinyl imidazole and tosylated CD. The resulting hybrid system, CNT-P, was then used as a support for the immobilization of Pd nanoparticles and furnishing a heterogeneous catalyst, Pd@CNT-P, with the utility for the hydrogenation of nitroarenes in aqueous media. The catalyst exhibited high catalytic activity and selectivity towards hydrogenation of nitro group. Moreover, it was recyclable up to six reaction runs. Comparing the catalytic activity of the catalyst with that of some control catalysts, the contribution of CD-IL and silica coated CNT to the catalysis has been confirmed. It was believed that CD in the backbone of the catalyst could act as a phase transfer agent, while multiple functional groups on the polymeric moiety improved Pd anchoring and suppressed its leaching. An efficient CAN-mediated C-C bond formation in the oxidative reactions of xanthenes with enaminones under mild reaction conditions are described. Dementia is a cognitive disorder mostly associated with Alzheimer’s disease (AD) in addition to being seen in many other diseases of the central nervous system (CNS). The limited number of drugs is not sufficient to provide adequate improvement to increase the quality of life of patients suffering from this symptom; therefore, all treatment options should be evaluated in detail. In this study, new molecules, [2-(4-(2/3/4-substituted phenyl)piperazin-1-yl)-4-phenylthiazol-5-yl][3/4-substituted phenyl]methanone derivatives (1-44), were obtained and analyzed in terms of their anticholinesterase activities. Kinetic mode and molecular interactions were also evaluated. An enzyme inhibition study was undertaken on acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) using the Ellman method. Maestro program was used in molecular modeling studies. Forty-four compounds were evaluated on AChE and BChE enzymes at 10(-3) and 10(-4) concentrations. The inhibition concentrations were calculated as 0.268 mu M to 2.104 mu M for six compounds (4, 5, 16, 27, 37, and 38) on AChE. Compound 5 including the 4-methoxy substituent (IC50: 0.268 mu M) and compound 38 containing the 4-methoxy and 3-methyl substituents (IC50: 0.286 mu M) showed the highest AChE inhibitory activity. They were further examined in terms of hydrogen bonding with Arg296 and Ar-Ar interaction with Trp286. The activity of compound 5 was also assessed in mixed-type kinetic mode.
