The aim of the present study is to design and synthesis of new pyrazole-triazolopyrimidine hybrids as potent alpha-glucosidase inhibitors. The target compounds 4a-n were synthesized by one-pot multicomponent approach with good yields and were characterized by various spectroscopic techniques and finally by single crystal X-ray diffraction method (4j). All the newly-synthesized derivatives have been screened for their alpha-glucosidase enzyme inhibition activity and acarbose taken as a standard drug. Among all the tested compounds, 4h has displayed excellent alpha-glucosidase enzyme inhibition activity with IC50 value 12.45 mu M to the standard drug acarbose (IC50: 12.68 mu M). Similarly, the compounds 4f and 4l have exhibited potent activity with IC50 values 14.47 mu M and 17.27 mu M respectively. Structure-activity relationship (SAR) studies of all the title compounds were established. The mode of binding interactions between the alpha-glucosidase enzyme and the compounds were studied. The drug-likeness properties (Lipinski parameters and in silico ADME properties) have predicted for the target compounds. The alpha-glucosidase inhibition, molecular docking and drug-likeness properties of the compounds 4h, 4f and 4l were suggested that these are promising hits for anti-diabetic activity.

A new Au(I)-catalyzed method for the preparation of trisubstituted indolizines from easily accessible 2-propargyloxy-pyridines is reported. The reaction tolerates a wide range of functionality, allowing for diversity to be introduced in four distinct regions of the product (R, R-1, R-2, and Ar). The reduction of graphene oxide (GO) was examined by using the extracts of different plants; cloves, white mulberry, black cumin seed, blackthorn, dark grape, and rosehip. The reduction efficiency of extracts was confirmed by XRD and spectroscopic analysis. The layered structure of reduced graphene and graphene nanocomposite decorated with palladium nanoparticles (PdNPs) was observed by electron microscopy techniques. This nanocomposite considered to be an active-selective catalyst for hydrogenation of nitroarenes in organic synthesis. This efficient catalyst can provide to develop clean, environmentally friendly, and energy-efficient production routes in industrial organic chemistry. It was concluded that the PdNPs/RGO catalyst showed high stability based on its reusing performance; seven times without loss of catalytic activity. (C) 2019 Hydrogen Energy Publications LLC. o-Phenylenediammines ando-aminophenols were reacted with alpha-oxodithioesters in a highly regioselective fashion to give 2-methylthio-3-aryl/heteroarylquinoxalines and 2-acylbenzoxazoles in 55-94% and 45-86%, respectively, in the presence ofp-toluene sulfonic acid catalyst. Control experiments involving reaction of aniline with a alpha-oxodithioester indicated that the thiocarbonyl group is more reactive than the carbonyl group. Based on this, probable mechanisms for the formation of quinoxalines and benzoxazoles are given. Biological targets of the quinoxalines and benzoxazoles were identified by bioinformatics. It was found that quinoxalines have good binding affinity with human dual-specificity tyrosine-phosphorylation-regulated kinase 1A and benzoxazoles with human carboxylesterase.

Selective hydrogenation of nitro compounds requires highly active and less expensive materials as a heterogeneous catalyst. As a good candidate, the Ni-Au alloy nanoparticles (NPs) have been synthesized by simple in-situ reduction method, which was loaded to the active carbon with 10 wt % loading amount. The Ni-Au/C catalyst shows superior catalytic performance for hydrogenation of nitro compounds with 100% conversion and selectivity without any by-products, which also indicates good recycle performance for several times use. The prepared catalyst not only reduces the usage amount of noble metal, but also keeps the high catalytic activity, which is a promising catalyst for industry applications. (C) 2019 Elsevier B.V. Various 2-aryl-3-vinyl-indoles were generated in situ through dehydrative condensation of aryl ketones and indoles. In view of expanding the structure activity relationship of xanthine oxidase inhibitors, a series of 3-oxo-6-aryl-2,3-dihydropyridazine-4-carbohydrazide/carboxylic acid derivatives were designed by molecular docking and synthesized. All the target compounds were evaluated for their in vitro XO inhibition by using febuxostat and allopurinol as the standard controls. Most of the hydrazide derivatives exhibited potency levels in the micromolar range. From the view of docking study, hydrazide derivatives bind to the active site of XO through a novel interaction mode, which is different from that of febuxostat bearing a carboxyl group. The most promising compound 8b was further subjected to kinetic analysis to deduce their modes of inhibition.
