4.1.1.1. 6-Bromo-3-((2,3-dihydrobenzo[b][1,4]dioxin-5-yl)methyl)-2-methoxyquinoline (VI: Y = 2,3-O(CH2)2O-). To a solution of 2,3-dihydrobenzo[b][1,4]dioxine-5-carboxylic acid (5.00 g, 28.0 mmol)in THF (150 mL) at 0 C was added lithium aluminium hydride(2.13 g, 56.0 mmol) in small portions. The reaction mixture wasstirred at 0 C for 10 min and stirred for a further 18 h at 20 C.Water (150 mL) was added to the reaction mixture which wasextracted with EtOAc (2 100 mL). The combined organic layerswere washed with brine (100 mL), dried over Na2SO4, filteredand concentrated under reduced pressure to obtain (2,3-dihydrobenzo[b][1,4]dioxin-5-yl)methanol as a yellow oil (3.22 g,99%). 1H NMR (CDCl3, 400 MHz) d 6.87-6.79 (m, 3H), 4.66 (s, 2H),4.32-4.30 (m, 2H), 4.28-4.25 (m, 2H), 2.19 (bs, 1H). Found: [M+H-18] = 149.5.

Example 8 Synthesis of 4-{2-[(2,3-Dihydro-benzo[1,4]dioxine-5-carbonyl)-amino]-benzooxazol-6-yloxy}-pyridine-2-carboxylic acid methylamide (Table 2, Compound 110) 4-(2-Amino-benzooxazol-6-yloxy)-pyridine-2-carboxylic acid methylamide (1 eq) and 2,3-dihydro-1,4-benzodioxane-5-carboxylic acid (1 eq) were dissolved in DMF. To this solution were added DIPEA (3 eq) and [dimethylamino-([1,2,3]triazolo[4,5-b]pyridin-3-yloxy)-methylene]-dimethyl-ammonium hexafluoro phosphate (1 eq). The mixture was stirred at 40¡ã C. overnight, then diluted with ethyl acetate and washed once each with 1N HCl, saturated aqueous sodium bicarbonate, and brine, and finally dried with anhydrous sodium sulfate, filtered, and concentrated. This was purified by preparatory reverse phase HPLC. MH+=447.0.

Method I Ba (1 mmol) and 2,3-dihydrobenzo[b][1,4]dioxin-6-carboxylic acid (or 2,3-dihydrobenzo[b][1,4]dioxin-2-carboxylic acid) (1 mmol) together with EDCI (1.5 mmol) and HOBt (0.05 mmol) in CH2Cl2 (20 mL) were refluxed at room temperature for 10 h. While the reaction completed, the solution was washed with water for three times (30 mL each time). The remaining water layer was extracted by EtOAc for three times (30 mL each time). The organic layers (CH2Cl2 and EtOAc) were combined and then evaporated. EXAMPLE 22 Preparation of (-)-N-[[3-[4-[4-[(2,3-Dihydro-1,4-benzodioxin-2-yl)carbonyl]-1-piperazinyl]-3-fluorophenyl]-4,5-dihydro-5-isoxazolyl]methyl]acetamide. To a flame dried flask containing (-)-N-[[4,5-dihydro-3-[3-fluoro-4-(1-piperazinyl)phenyl]-5-isoxazolyl]methyl]acetamide (Step 1, Example 21) (200 mg), 1,4-benzodioxan-2-carboxylic acid (135 mg), DMAP (9 mg) in pyridine (7 mL) is added EDC (144 mg) at ambient temperature and stirred for 48 hours. The reaction is diluted with CH2Cl2 (25 mL) and washed with H2O (2 X 50 mL) and saline (50 mL). The organic phase is dried over sodium sulfate, concentrated in vacuo and chromatographed on silica gel (230-400 mesh), eluding with chloroform/methanol (99/1). The appropriate fractions are combined (Rf= 0.29,TLC, chloroform/methanol, 95/5), concentrated to give the title compound, mp 158-161 ¡ãC, [alpha]25D -43¡ã (CHCl3)., 3663-80-7

Step 1..Preparation of 3-((2,3-dihydrobenzo[b][1 ,4]dioxin-6-yl)methyl)-2-imidazolidin-4- one; A suspension of (2,3-Dihydro-benzo[1 ,4]dioxin-6-yl)-methylamine (16.5 mmol, 2.73 g) in 1 MNaOH is refluxed for 10 min and cooled to room temperature. Subsequently bis(carboxymethyl) trithiocarbonate (3.74 g, 16.5 mmol) is added and the reaction mixture refluxed overnight to give the precipitate, which is filtered off and is washed with water (3x 20 ml_). The remaining residue is suspended in a mixture of methanol and isopropanol (1 :1 , 25 ml_) and refluxed for 10 minutes. Cooled reaction mixture is filtered and the solid residue dried at lower temperature and recrystallyzed from methanol to give 3.29 g of 3-((2,3- dihydrobenzo[b][1 ,4]dioxin-6-yl)methyl)-imidazolidin-2,4-dione.
