Preparation of 2,2-disubstituted and 2-monosubstituted alkenylboronic acid esters from aliphatic and aromatic ketones and aldehydes by the boron-Wittig olefination with bis[(pinacolato)boryl]methane was examined and applied on the multigram scale. The influence of the substrate steric and electronic features on the overall efficiency and stereochemical outcome of the reaction was studied. Additionally, a series of diversely functionalized (hetera)cycloalkylidenemethyl and (hetera)cycloalkyl boronic acid-derived building blocks was synthesized.

The objective of this work is to apply yellow mustard seeds in the enzymatic reduction (biocatalysis) of acetophenone and its derivatives, providing a new biocatalyst for this type of reduction. The reaction parameters were optimized from acetophenone, resulting in 1-phenylethanol, with 68.5% conversion yield and 53% enantiomeric excess (e.e.), analyzed by GC and HPLC, respectively. Reactions to acetophenone and its derivatives were conducted in aqueous medium using buffers pH 6.0 and pH 6.5 in the presence of PVP. A polymer that can act as a protector preventing oxidation of peroxidase enzymes. The results for the derivatives presented low yields (4-18%: substances 5, 7 and 8), moderate (28-40%: substances 3, 4 and 6) and optimum (81%: substance 2) yields, with enantiomeric excesses varying from good to excellent results. Meta-substituted derivatives showed inversion of configuration in the presence of PVP. The biocatalyst was also tested in reactions with other carbonyl compounds, presenting moderate results. In this way, this work offers a new perspective of the application of PVP in biocatalysis reactions and a new biocatalyst for future applications.

The synthesis of previously unattainable 2,5-disubstituted 4,5-dihydroimidazo[1,5-b]pyrazol-6-ones has been developed. Electrochemical reductions of readily available 2,2,2-trichloroethylideneacetophenones were followed by reaction with hydrazine, leading to 3-aryl-5-dichloromethyl-2-pyrazolines. These were treated with isocyanates to obtain the corresponding aminocarbonyl derivatives, which were found to be able to form an otherwise almost inaccessible imidazo[1,5-b]pyrazole ring system via a one-step reaction involving internal condensation followed by hydrogen chloride elimination and aromatization. The molecular- structures of 2-(4-methylphenyl)-5-tosyl-4,5-dihydro-imidazo[1,5-b]pyrazol-6-one, 5-dichloromethyl-N-(4-chlorophenyl)-4,5-dihydro-3-p-tolylpyrazole-1-carboxamide, and 5-(4-bromophenyl)-2-p-tolyl-4,5-dihydroimidazo[1,5-b]pyrazol-6-one were determined by X-ray crystallographic analysis.

We report an efficient HCl center dot DMPU assisted one-pot conversion of aldehydes into nitriles. The use of HCl center dot DMPU as both an acidic source as well as a non-nucleophilic base constitutes an environmentally mild alternative for the preparation of nitriles. Our protocol proceeds smoothly without the use of toxic reagents and metal catalysts. Diverse functionalized aromatic, aliphatic and allylic aldehydes incorporating various functional groups were successfully converted to nitriles in excellent to quantitative yields. This protocol is characterized by a broad substrate scope, mild reaction conditions, and high scalability.

A simple and practical domino protocol for the selective synthesis of 2-aroylbenzothiazoles and 2-aryl benzothiazoles catalyzed by I-2 is developed under metal-free conditions. The reaction outcomes are exclusively controlled by the reaction oxidant/medium. With DMSO employed as both the solvent and the oxidant, an oxidation of aromatic methyl ketones takes precedence over the condensation with 2-aminobenzenethiols. On the other hand, when the reaction was carried out in PhNO2 or in 1,4-dioxane containing PhNO2, the condensation of aromatic methyl ketones with 2-aminobenzenethiols has priority to form imines which is followed by an oxidation of the methyl group from ketones to afford 2-arylbenzothiazoles as a sole product. The PhNO2/I-2 co-catalytic system is proposed first time.

Catalytic hydrogenation of nitroaromatics is an environment-benign strategy to produce industrially important aniline intermediates. Herein, we report that Fe(OH)(x) deposition on Pt nanocrystals to give Fe(OH)(x)/Pt, enables the selective hydrogenation of nitro groups into amino groups without hydrogenating other functional groups on the aromatic ring. The unique catalytic behavior is identified to be associated with the Fe-III-OH-Pt interfaces. While H-2 activation occurs on exposed Pt atoms to ensure the high activity, the high selectivity towards the production of substituted aniline originates from the Fe-III-OH-Pt interfaces. In situ IR, X-ray photoelectron spectroscopy (XPS), and isotope effect studies reveal that the Fe3+/Fe2+ redox couple facilitates the hydrodeoxygenation of the -NO2 group during hydrogenation catalysis. Benefitting from Fe-III-OH-Pt interfaces, the Fe(OH)(x)/Pt catalysts exhibit high catalytic performance towards a broad range of substituted nitroarenes.

A new rhodium(III) triphenylphosphine complex having the general formula [Rh(PPh3)(2)(L)Cl] (1) was synthesized by C-S bond cleavage of an ONS donor azo-thioether ligand (L-CH2Ph). The complex was thoroughly characterized by various spectroscopic techniques. Its single crystal X-ray structure exhibits an octahedral geometry around the rhodium(III) center. A cyclic voltammogram of the complex exhibits ligand based quasi-irreversible oxidative and reductive responses. The electronic structure, redox properties and electronic excitations in the complex were interpreted by DFT and TDDFT calculations. Herein, the composite of palladium nanoparticles and graphene oxide (CPG) has been synthesized by a facile and very efficient method that provided chemical selectivity and high catalytic activity. The synthesized CPG was characterized by several techniques such as transmission and high-resolution electron microscopy (TEM and HR-TEM), X-ray diffraction (XRD) and Raman spectroscopy, and Photoelectron spectroscopy (XPS). CPG was tested for selective reduction of nitroarenes at room temperature. After the addition of CPG to the reaction media, catalytic performances were depended upon the cooperative effect of hydrogen activation with Pd nanoparticles, where the lack of electrons favors an excellent performance. Nitroarenes can be bound to the energetically preferred adsorption site for the nitro group in electrically enriched graphene oxide. In addition, the Pd nanoparticles transfer electrons to the graphene oxide which increases the functions of metal and carbon support. CPG exhibits both chemoprotective and high catalytic performance for hydrogenation of nitroarenes at room temperature. Aniline derivatives were obtained with high yields under mild conditions and a practical catalytic system was developed by the use of CPG.

An efficient synthesis of pyrimidines and pyridines has been developed from readily available aromatic ketones, aldehydes and ammonium salts under transition-metal-free conditions. In this strategy, ammonium salts were used as nitrogen sources and only water was generated as a nontoxic byproduct. A catalytic amount of NaIO4 played an important role in the selectivity control, whereas substituted pyridines were dominantly formed in its absence.

Many natural or synthetic chalcones have potential anti-tumor activity. Here, we synthesized two series of chalcone analogues containing a thieno [2,3-cflpyrimidin-2-y] group and evaluated for their cytotoxic activity towards cultured human lung cancer A549 and colorectal HCT-116 cells. Among them, compound 8d was the most cytotoxic against HCT-116 cells, with an IC50 value of 2.65 mu M. Analyses of the phenotypic changes induced by this compound found a dose-dependent accumulation of HCT-116 cells in sub-G1 phase, indicating that compound 8d might induce apoptosis. Furthermore, we found that 8d triggered mitochondrial membrane potential depolarization, promoted reactive oxygen species formation in HCT-116 cells, and increased the percentage of early and late apoptotic cells. Finally, immunoblotting indicated that 8d increased PARP-1 and caspases 3, 7 and 9 cleavage. These data suggest that compound 8d induces apoptosis via the mitochondrial death pathway.
