DDIBNPPI and DDIBPPIN were obtained by rupturing the covalent bond of phenanthrimidazole core to prevent aggregation-induced quenching. The DDIPNPPI and DDIBPPIN materials employed as a host to fabricate green and red phosphorescent organic light-emitting diodes. The red/white devices (with 0.4% dopant concentration) with DDIBPPIN:Ir(MDQ)2(acac) exhibit maximum L of 69889/26319 cd/m2, etaex of 19.6/16.6%, etac of 34.6/35.6 cd/A, and etap of 35.8/36.6 lm/W. The device with DDIBPPIN:Ir(ppy)3/DDIPNPPI:Ir(ppy)3 exhibits green emission [Commission Internationale de l’Eclairage (0.30,0.60)/(0.30,0.60)] with maximum L of 69906/69482 cd/m2, etaex of 17.9/17.0%, etac of 59.8/58.6 cd/A, and etap of 63.6/61.3 lm/W. The photophysical properties of the synthesized 4-amino-and 4-acetylaminonaphthalimides depend on the solvent polarity and its specific solvating ability due to H-bonding. The vesicular glutamate transport (VGLUT) system selectively mediates the uptake of L-glutamate into synaptic vesicles. Uptake is linked to an H+-ATPase that provides coupling among ATP hydrolysis, an electrochemical proton gradient, and glutamate transport. Substituted quinoline-2,4-dicarboxylic acids (QDCs), prepared by condensation of dimethyl ketoglutaconate (DKG) with substituted anilines and subsequent hydrolysis, were investigated as potential VGLUT inhibitors in synaptic vesicles. A brief panel of substituted QDCs was previously reported (Carrigan et al. Bioorg. Med. Chem. Lett. 1999, 9, 2607-2612)1 and showed that certain substituents led to more potent competitive inhibitors of VGLUT. Using these compounds as leads, an expanded series of QDC analogues were prepared either by condensation of DKG with novel anilines or via aryl-coupling (Suzuki or Heck) to dimethyl 6-bromoquinolinedicarboxylate. From the panel of almost 50 substituted QDCs tested as inhibitors of the VGLUT system, the 6-PhCH=CH-QDC (Ki = 167 muM), 6-PhCH2CH2-QDC (Ki = 143 muM), 6-(4?-phenylstyryl)-QDC (Ki = 64 muM), and 6-biphenyl-4-yl-QDC (Ki=41 muM) were found to be the most potent blockers. A preliminary assessment of the key elements needed for binding to the VGLUT protein based on the structure-activity relationships for the panel of substituted QDCs is discussed herein. The substituted QDCs represent the first synthetically derived VGLUT inhibitors and are promising templates for the development of selective transporter inhibitors.

Liver X receptor (LXR) is a member of the nuclear receptor superfamily, and it regulates various biologic processes, including de novo lipogenesis, cholesterol metabolism, and inflammation. Selective inhibition of LXR may aid the treatment of nonalcoholic 1fatty liver diseases. In the present study, we evaluated the effects of three cinnamamide derivatives on ligand-induced LXRalpha activation and explored whether these derivatives could attenuate steatosis in mice. N-(4-trifluoromethylphenyl) 3,4-dimethoxycinnamamide (TFCA) decreased the luciferase activity in LXRE-tk-Luc-transfected cells and also suppressed ligand-induced lipid accumulation and expression of the lipogenic genes in murine hepatocytes. Furthermore, it significantly attenuated hepatic 1neutral lipid accumulation in a ligand-induced fatty liver mouse 1system. Modeling study indicated that TFCA inhibited activation of the LXRalpha ligand-binding domain by hydrogen bonding to Arg305 in the H5 region of that domain. It regulated the transcriptional control exerted by LXRalpha by influencing coregulator exchange; this process involves dissociation of the thyroid hormone receptor-associated proteins (TRAP)/DRIP coactivator and recruitment of the nuclear receptor corepressor. These results show that TFCA has the potential to attenuate ligand-induced lipogenesis and fatty liver by selectively inhibiting LXRalpha in the liver.
