Novel vanilloid receptor-1 antagonists: 1. Conformationally restricted analogues of trans-cinnamides

The vanilloid receptor-1 (VR1 or TRPV1) is a member of the transient receptor potential (TRP) family of ion channels and plays a role as an integrator of multiple pain-producing stimuli. From a high-throughput screening assay, measuring calcium uptake in TRPV1-expressing cells, we identified an N-aryl trans-cinnamide (AMG9810, compound 9) that acts as a potent TRPV1 antagonist. We have demonstrated the antihyperalgesic properties of 9 in vivo and have also reported the discovery of novel, orally bioavailable cinnamides derived from this lead. Herein, we expand our investigations and describe the synthesis and biological evaluation of a series of conformationally constrained analogues of the s-cis conformer of compound 9. These investigations resulted in the identification of 4-amino- and 4-oxopyrimidine cores as suitable isosteric replacements for the trans-acrylamide moiety. The best examples from this series, pyrimidines 79 and 74, were orally bioavailable and exhibited potent antagonism of both rat (IC50 = 4.5 and 0.6 nM, respectively) and human TRPV1 (IC50 = 7.4 and 3.7 nM, respectively). In addition, compound 74 was shown to be efficacious at blocking a TRPV1-mediated physiological response in vivo in the capsaicin-induced hypothermia model in rats; however, it was ineffective at preventing thermal hyperalgesia induced by complete Freund’s adjuvant in rats.

The regioselective catalytic synthesis of substituted quinoline and quinoxaline derivatives was achieved from the ortho-C-H bond activation reaction of arylamines and terminal alkynes by using the catalyst Ru3-(CO)12/HBF 4¡¤OEt2. The normal isotope effect (k CH/kCD = 2.5) was observed for the reaction of C 6H5NH2 and C6D5NH 2 with propyne. A highly negative Hammett value (rho = -4.4) was obtained from the correlation of the relative rates from a series of meta-substituted anilines, m-XC6H4NH2, with sigmap in the presence of Ru3(CO)12/ HBF4¡¤OEt2 (3 mol % Ru, 1:3 molar ratio). The deuterium labeling studies from the reactions of both indoline and acyclic arylamines with DC?CPh showed that the alkyne C-H bond activation step is reversible. The crossover experiment from the reaction of 1-(2-amino-1-phenyl)pyrrole with DC?CPh and HC?CC6H4-p-OMe led to preferential deuterium incorporation to the phenyl-substituted quinoline product. A mechanism involving rate-determining ortho-C-H bond activation and intramolecular C-N bond formation steps via an unsaturated cationic ruthenium acetylide complex has been proposed.

Achiral pyran-2-one analogs possessing a 3-S-(2-alkylphenyl) group were determined to be high-affinity inhibitors for human immunodeficiency virus (HIV) protease (PR). Crystallographic, modeling, and structure-activity studies led to the 3-S-(2-tert-butylphenyl) moiety as an apparent optimal group to access the S2?/S1? pockets of the enzyme. Further optimization led to an inhibitor, 3-[(2-tert-butylphenyl)sulfanyl]-4-hydroxy-6-(3-methylphenyl)-pyran-2-one (14), possessing a Ki of 3 nM. An X-ray crystallographic structure of an inhibitor, 4-hydroxy-3-[(2-isopropylphenyl)sulfanyl]-6-phenylpyran-2-one (8), bound to HIV PR showed that the 3-S-(2-isopropylphenyl) group occupied the P2? and P1? pockets, while other crucial interactions were common to those found with other pyran-2-one analogs. The high potency observed for this series may be due, in part, to the restrictions on the intramolecular collapsibility of these molecules in aqueous solution, leading to a highly favorable hydrophobic effect on binding. Herein we report a novel P2?/P1? achiral ligand which results in a tight-binding inhibitor that occupies only three pockets in the enzyme and exhibits a unique pattern of in vitro resistance.

A Redox Isomerization Strategy for Accessing Modular Azobenzene Photoswitches with near Quantitative Bidirectional Photoconversion

Photoswitches capable of accessing two geometric states are highly desirable, especially if their design is modular and incorporates a pharmacophore tethering site. We describe a redox isomerization strategy for synthesizing p-formylazobenzenes from p-nitrobenzyl alcohol. The resulting azo-aldehydes can be readily converted to photoswitchable compounds with excellent photophysical properties using simple hydrazide click chemistry. As a proof of principle, we synthesized a photoswitchable surfactant enabling the photocontrol of an emulsion with exceptionally high spatiotemporal precision.
