The synthesis of 1,2,3-triazoles with a sulfur-based side chain has been accessed with the metal-free annulation reactions of readily available beta-thiolated enaminones and tosyl hydrazine. By these reactions with water as the only medium, a broad array of 5-thiolated 1,2,3-triazoles have been synthesized with generally good to excellent yields. Except using TMEDA (N,N,N’,N’-tetramethylethylenediamine) as the only base promoter, not any other catalyst or additive is required, thus providing an efficient and environmentally benign method for useful 1,2,3-triazole synthesis.

In this study, a new heterobimetallic Mo(VI)-V(V) organosilicon Schiff base complex has been prepared and characterized by different techniques, such as FTIR, Raman, MS, ICP-AES, TGA, and XPS. The bimetallic nanocomplex, revealed by TEM images, showed high oxidation stability and desired activity in the aerobic oxidation of a structurally diverse set of benzylic alcohols in ethanol as a safe solvent. Further, oxidation of benzylic hydrocarbons successfully occurred, producing the target compounds in high yields and excellent selectivities. Our results demonstrated a cooperative effect between Mo(VI) and V(V) as redox active sites in an organosilicon Schiff base framework. A facile and practical reusability of the solid catalyst at the end of the reaction was observed.

The rearrangement of ene-aldimines is a useful reaction for affording homoallylic amines. Despite their utilities in synthetic chemistry, the rearrangement for accessing homoallylic amines substituted at the 2-position remains elusive. In this study, the Bronsted acid-initiated formal [1,3]-rearrangement of ene-aldimines was developed to synthesize 2,4,4-substituted homoallylic amines that were otherwise inaccessible previously. Our study reveals an intermolecular pathway in which the rearrangement proceeds via a protonation-mediated 2-azaallenium cation.

Novel enamine derivatives were synthesized from the reaction of lactone and chalcones and their antiproliferative and cytotoxic activities against six cancer cell lines (e. g., HeLa, HT29, A549, MCF7, PC3 and Hep3B) and one normal cell lines (e. g., FL) were investigated along with their mode of interactions with CT-DNA. Most of the enamine derivatives with IC(50)values of 86-168 mu M demonstrated much stronger antiproliferative activity than the starting molecules against the cancer cells. While, among the enamine derivatives, four compounds displayed higher cytotoxic potency than the control drugs (5-fluorouracil and cisplatin) against the Hep3B cell lines, these compounds did not exhibit any significant toxicity against normal cells, FL. The UV/VIS spectral data suggest that eight compounds cause hypochromism with a slight bathochromic shift (similar to 6 nm), indicating that they bind to the DNA by way of an intercalative or minor groove binding mode. The binding constants of the compounds are in the range of 0.1×103 M-1-2.3×104 M-1. The antiproliferative activity of studied enamine derivatives could possibly be due to their DNA binding as well as their cytotoxic properties. In addition to these assays, the chalcones and enamine derivatives were investigated by molecular docking to calculate the synergistic effect of antiproliferative activities against six human cancer cell lines.

A facile and general synthesis of 2,4,6-triaryl pyrimidines has been developed. It involves a one-pot [2+1+1+1+1] pseudo five-component annulation of one methyl ketone, two aldehydes and two NH4OAc catalyzed by TfOH. One C-C and four C-N bonds are formed during the oxidative annulation process. The reaction shows good tolerance of many important functional groups in air and produces only water as the coproduct, making this methodology a highly versatile alternative to the existing methods for structuring pyrimidine framework.

Ras proteins are small GTPases which regulate cellular proliferation, differentiation, and apoptosis. Constitutively active mutant Ras are expressed in similar to 15-20% human cancers, and K-Ras mutations account for similar to 85% of all Ras mutations. Since K-Ras must interact with the plasma membrane (PM) for biological activity, inhibition of the K-Ras/PM interaction is a tractable approach to block oncogenic K-Ras activity. Here, we discovered chalcones 1 and 8 exhibit anti-K-Ras activity, and show that the compounds mislocalize K-Ras from the PM and block oncogenic K-Ras signal output. Also, 1 inhibits the growth of K-Ras-driven human cancer cells. Our data suggest that 1 could be a promising starting point for developing anti-K-Ras cancer drug.

This work presents a highly active and reusable heterogeneous film catalytic assembly for hydrogenation reduction of aromatic nitro compounds. The multilayer structures of PEI-(K2PdCl4-P1)(n)-film (PEI = polyethylenmine, P1 = 3-amino-3-(4-pyridinyl)-propionitrile) bound to quartz slides were fabricated by layer-by-layer (LbL) self-assembly method. Various characterization techniques including X-ray photoelectron spectroscopy (XPS), inductively coupled plasma OES spectrometer (ICP), UV-vis spectroscopy and atomic force microscopy (AFM) were employed to reveal the growth process of the resulting LbL multilayers in detail. Subsequent in situ reduction by H-2 produced Pd nanoparticles embedded in such films were used as catalyst for the hydrogenation of nitroarenes. The catalytic performance test shows that the thin film catalyst can be applied to the hydrogenation reaction of various substituted nitroaromatics, and exhibits good catalytic activity and good catalyst stability. It is worth mentioning that our catalytic films can be easily removed from the reaction system in any time during the reaction, and the catalytic activity could be fully recovered when reused directly in another catalytic cycle for five times.

In the present investigation, an alluring green PEG-400 mediated one-pot synthesis of novel 2-(2-hydrazinyl)thiazole derivatives from 1,2,6,7-tetrahydro-8H-indeno[5,4-b]furan-8-one has been unveiled via a novel synthetic pathway. The application of PEG in organic reactions as a reaction media is one of the fantastic tools of green chemistry as reactions can be carried out under generous conditions limiting environmental peril and chemical waste. PEG-400 is recognized as a low-cost, non-flammable, environmentally benign, recyclable, and richly available green solvent. A series of novel 2-(2-hydrazineyl)thiazole derivatives have been synthesized in good to excellent yield by using the green capability of PEG-400 solvent. These newly synthesized compounds were tested for their antimicrobial and antioxidant activities. The results revealed that these compounds show good activities compared with the standard. Additionally, all the synthesized compounds exhibit negligible cytotoxicity as compared to the positive control. The structures of all novel compounds reported herein are established using FT-IR, H-1 NMR, C-13 NMR, D2O exchange experiment and HRMS techniques.
