A series of novel 1,2,3-triazole linked thiazole-1,2-isoxazole derivatives has been designed, synthesized and characterized by H-1 and C-13 NMR, and mass spectral analysis. The compounds have been tested for their anticancer activity towards MCF-7 (breast cancer), A549 (lung cancer), Colo-205 (colon cancer), and A2780 (ovarian cancer) by using the MTT method using etoposide as the reference. Most of the tested compounds demonstrate good to moderate activity against all cell lines. The compounds 14b, 14e, 14g, and 14h are characterized by inhibitory activity stronger than that of etoposide.

Peroxynitrite (ONOO-), a highly reactive species, is profoundly involved in many physiological and pathological processes. Change of the ONOO- level usually indicates an abnormal body function. Thus, it is desired to develop a highly reliable ONOO- assay to elucidate its roles in a related disease environment. In this work, we have constructed a ratiometric molecule fluorescent probe RTFP toward ONOO- with high specificity by the combination strategy of probe screening and a rational design method. RTFP displayed excellent detection sensitivity (detection limit: 4.1 nM) and produced a highly ratiometric emission signal (130-fold). Leveraging this probe, we showed the change of ONOO- content in the free-fatty-acid-induced nonalcoholic fatty liver disease (NAFLD) and acetaminophen-induced drug-induced liver injury (DILI) cellular model and for the first time disclosed the involved mechanism of cytochrome P450 2E1 (CYP2E1) enzyme in NAFLD with a DILI pathological environment. Furthermore, RTFP also was utilized to visualize ONOO- fluctuation of living liver tissues in a high-fat-diet-caused NAFLD model. We expected that this probe may help the study of liver injury in the exploration of mechanism and signal path.

Y By removing 5-methyl and 6-acetyl groups in our previously reported compound 3, we designed a series of novel 2,7-diaryl-[1,2,4]triazolo[1,5-a]pyrimidine derivatives as potential tubulin polymerization inhibitors. Among them, compound 5e displayed low nanomolar antiproliferative efficacy on HeLa cells which was 166-fold higher than the lead analogue 3. Interestingly, 5e displayed significant selectivity in inhibiting cancer cells over HEK-293 (normal human embryonic kidney cells). In addition, 5e dose-dependently arrested HeLa in G2/M phase through the alterations of the expression levels of p-cdc2 and cyclin B1, and caused HeLa cells apoptosis by regulation of expressions of cleaved PARP. Further evidence demonstrated that 5e effectively inhibited tubulin polymerization and was 3-fold more powerful than positive control CA-4. Moreover, molecular docking analysis indicated that 5e overlapped well with CA-4 in the colchicine-binding site. These studies demonstrated that 2,7-diaryl-[1,2,4]triazolo [1,5-a]pyrimidine skeleton might be used as the leading unit to develop novel tubulin polymerization inhibitors as potential anticancer agents. The use of ubiquities elements such as iron instead of expensive precious metals as catalysts is one goal toward realizing environmentally benign synthetic chemistry. Here, we report that porous FeO(OH) dispersed on Mg-Al hydrotalcite acts as a bifunctional heterogeneous catalyst in the one-pot synthesis of 2-substituted quinoline derivatives through dehydrogenative oxidation-cyclization reactions. The catalyst was prepared by a simple grafting method using FeCl3 and Mg-Al hydrotalcite. The prepared porous FeO(OH) possesses a higher surface area than those previously reported for alpha-FeO(OH) particles. The one-pot quinoline synthesis proceeded effectively under non-noble-metal catalysis in air without requiring additional homogeneous bases or solvents.

Zr-based metal-organic frameworks have been considered promising heterogeneous catalysts in various reactions. It has been proven that the defect sites in these frameworks, which result from deficiency of linkers, could act as the reaction active sites. In this work, microwave irradiation was used for rapid synthesis and increasing defects on Zr-6 clusters of metal-organic framework UiO-66. Powder X-ray diffraction, thermal gravimetric analysis, and N-2 sorption isotherms were performed to characterize and calculate the defect number in the framework. UiO-66 prepared by microwave method for a few minutes could reach 1.8 missing linkers per cluster. The material with acetic acid as a co-catalyst showed highly heterogeneous catalytic activity in the solvent-free heterocyclization of 2,2-disubstituted 2,3-dihydroquinazolin-4(1H)-ones from anthranilamide and ketones/aldehydes. The synthetic scheme consisted of zirconium cluster catalysed carbonyl group of anthranilamide to achieve the C-N propitious combination and succeeding ring closure process.

Switchable selective hydrogenation among the groups in multifunctional compounds is challenging because selective hydrogenation is of great interest in the synthesis of fine chemicals and pharmaceuticals as a result of the importance of key intermediates. Herein, we report a new approach to highly selectively (>99%) reducing C=X (X = O, N) over the thermodynamically more favorable nitro groups locating the substrate in a metal-organic capsule containing NADH active sites. Within the capsule, the NADH active sites reduce the double bonds via a typical 2e(-) hydride transfer hydrogenation, and the formed excited-state NAD(+) mimics oxidize the reductant via two consecutive 1e(-) processes to regenerate the NADH active sites under illumination. Outside the capsule, nitro groups are highly selectively reduced through a typical 1e(-) hydrogenation. By combining photoinduced 1e(-) transfer regeneration outside the cage, both 1e(-) and 2e(-) hydrogenation can be switched controllably by varying the concentrations of the substrates and the redox potential of electron donors. This promising alternative approach, which could proceed under mild reaction conditions and use easy-to-handle hydrogen donors with enhanced high selectivity toward different groups, is based on the localization and differentiation of the 2e(-) and 1e(-) hydrogenation pathways inside and outside the capsules, provides a deep comprehension of photocatalytic microscopic reaction processes, and will allow the design and optimization of catalysts. We demonstrate the advantage of this method over typical hydrogenation that involves specific activation via well-modified catalytic sites and present results on the high, well-controlled, and switchable selectivity for the hydrogenation of a variety of substituted and bifunctional aldehydes, ketones, and imines.

With the aim to discover novel, efficient and selective inhibitors of human alkaline phosphatase and nucleotide pyrophosphatase enzymes, two new series of pyrazolyl pyrimidinetriones (PPTs) (6a-g) and thioxopyrimidinediones (PTPs) (6h-n) were synthesized in good chemical yields using Knoevenagel condensation reaction between pyrazole carbaldehydes (4a-g) and pharmacologically active N-alkylated pyrimidinetrione (5a) and thioxopyrimidinedione (5b). The inhibition potential of the synthesized hybrid compounds was evaluated against human alkaline phosphatase (h-TNAP and h-IAP) and ectonucleotidase (h-NPP1 and h-NPP3) enzymes. Most of the tested analogs were highly potent with a variable degree of inhibition depending on the functionalized hybrid structure. The detailed structure-activity relationship (SAR) of PPT and PTP derivatives suggested that the compound with unsubstituted phenyl ring from PPT series led to selective and potent inhibition (6a; IC50= 0.33 +/- 0.02 mu M) of h-TNAP, whereas compound 6c selectively inhibited h-IAP isozyme with IC50 value of 0.86 +/- 0.04 mu M. Similarly, compounds 6b and 6h were identified as the lead scaffolds against h-NPP1 and h-NPP3, respectively. The probable binding modes for the most potent inhibitors were elucidated through molecular docking analysis. Structure-activity relationships, mechanism of action, cytotoxic effects and druglikeness properties are also discussed.
